Bu çalışmada, Türkiye'nin farklı coğrafik bölgelerinden kaçak ekimler sonucu ele geçirilerek, İstanbul Adli Tıp Kurumu tarafından fakültemize teslim edilen kenevir tohumları arasında akrabalık bulunup bulunmadığı RAPD dominant markörleri kullanılarak araştırılmıştır. Dominant markörlerin istatistiki analizlerinde kullanılan temel koordinatlar analizi, kümeleme analizi, kümelerin güven sınırları, Mantel testi ve Nei ve Li genetik mesafesi tanıtılmış ve elde edilen veriler üzerinde bu metotlar uygulanmıştır. Bu çalışmada 29 kenevir aksesyonu sera saksı ortamında DNA izolasyonu için yaprak örnekleri alınıncaya kadar yetiştirilmiştir. Moleküler genetik çalışmalarda tek birey DNA (SET1) ve bulk DNA (SET2) setleri kullanılmıştır. PCR amplifikasyonları sonucunda SET1'de 264, SET2'de 241 DNA bantı skorlanmıştır. Elde edilen verilerin istatistiki analizleri sonucunda SET1'de kullanılan aksesyonlar arasında belirgin bir gruplaşma gözlenmemiştir. SET2'de ise ülkemizin batısındaki illerden elde edilen kenevir aksesyonları ile, doğusundaki illerden elde edilen kenevir aksesyonlarının iki farklı grup oluşturduğu gözlenmiştir.
In this study, Cannabis Sativa L. accessions which were seized from different parts of Turkey were used. The Istanbul forensic institute provided the Cannabis accessions, which used in this study. Aim of the study was to determine genetic relatedness between the accessions. For this purpose dominant markers (RAPD) were employed. In addition, statistical methods (such as principal coordinate analysis, cluster analysis, bootstraps Mantel test, and Nei and Li genetic distance), which are used in the analysis of dominant markers, were evaluated and employed in the analysis of Cannabis data. In this study 29 Cannabis accessions were grown in the green house conditions untill DNA extraction material obtained. In the molecular study, single Cannabis accession (SET1) and bulk of accessions of the same genotype (SET2) were used. PCR amplifications produced 264 markers in SET1 and 241 markers in SET2. Analysis of SET1 data did not show a separation between the accessions. On the other hand, when the SET2 data were analysed, Cannabis accessions were clearly separated into two main groups, one of which made up of Cannabis accessions which were seized from eastern part of Turkey while the other group was made up of Cannabis accessions which were seized from western part of Turkey
