Bu tez çalışmasında, gen merkezi Anadolu olan nohutun, kültür formları çok sınırlı bir genetik varyasyona sahip olduğu için, Anadolu'nun zengin yabani nohut biyoçeşitliliğinden yararlanılabilmesi maksadı ile bu türlerin genetik çeşitlilik ve benzerliklerinin RAPD ve ISSR teknikleri kullanılarak belirlenmesi hedeflenmiştir. RAPD ve ISSR markör teknikleri, aralarında kültürü yapılan Cicer arietinum'un da bulunduğu 7 tek yıllık Cicer türü genotiplerine uygulanmıştır. RAPD primerlerinden seçilen 14'ü, ISSR primerlerinden de yeterli polimorfik band üreten 16'sı kullanılarak toplam 385 band skorlanmıştır. NTSYS-pc, 2.0 programı kullanılarak elde edilen dendogramlar neticesinde tek yıllık Cicer türlerinin 2 ana kola ayrıldığı görülmüştür. İlk kümede kültür nohutu olan Cicer arietinum, Cicer reticulatum ve Cicer echinospermum, diğer kümede ise Cicer pinnatifidum, Cicer bijugum ve Cicer judaicum yer almıştır. Bu kollara uzak olan dış grupta ise Afganistan'da yetişen endemik bir tür olan Cicer yamashitae bulunmaktadır. Cicer reticulatum'un kültür nohutu Cicer arietinum'un yabani atası olduğu ve Türkiye'nin güneydoğusunun nohutun anavatanı olduğu savı çalışmamızın sonuçlarıyla da desteklenmiştir. Ayrıca, kültür nohutunun dar bir genetik havuzda yer aldığı, buna karşılık aynı melezleme grubunda yer alan C. reticulatum ve C. echinospermum'un genetik varyasyonunun geniş olduğu da analizlerimizle belirlenmiştir. Kültür nohutu ile direkt melezleme olanağı bulunmayan ikinci melezleme grubuna ait türlerin ise ilk gruptaki türlerden belirgin bir şekilde ayrıldığı gözlenmiştir.
This thesis study was conducted to reveal the wide genetic diversity of the Anatolian originated wild Cicer species in order to be used in the improvement programs of the narrow genetic pool of the chickpea cultivars. RAPDs and ISSRs were the molecular techniques employed to the 7 annual Cicer species, including the cultivar Cicer arietinum. Selected 14 RAPD primers were utilised together with 16 ISSRs and a total of 385 fragments were scored. NTSYS-pc version 2.0 was used to generate the dendograms that clearly classified the Anatolian Cicer species into two major groups. The first group was occupied by Cicer arietinum, C. reticulatum and C. echinospermum while in the second group C. pinnatifidum, C. bijugum and C. judaicum were included. C. yamashitae was an endemic plant of Afghanistan and was used as an outgroup. The hypothesis that C. reticulatum is the progenitor of chickpea and the southeastern part of Turkey is the place where this crop was first domesticated, was also supported with our study. Additionally, it was also determined that the cultivar has a narrow genetic pool while the wild relatives of the first crossability group, namely C. reticulatum and C. echinospermum, display a wide genetic variation. The wild Cicer species without the ability to be crossed with the cultivar, members of the second crossability group, were clearly seperated from the first group.
