Bu arastırmada, Antalya ilinde faaliyet gösteren bir firmaya ait parçalama ünitesinden ve marketlerden 12 ay süreyle ayrı ayrı olmak üzere antrekot, kontrnuar, yumurta, kontrfile, tranç sokum, gerdan, nuar ve bonfile et preperatları alındı. Her bir et numunesinden, 24 adet olmak üzere toplam 432 adet numune kullanıldı. Ayrıca marketlerden 60 adet kıyma numunesi temin edildi. Bütün numuneler, toplam mezofilik aerob, koliform bakterisi, E. coli ve koagulaz pozitif stafilokoklar yönünden incelenerek istatistiksel olarak degerlendirildi. Et parçalama ünitesinden temin edilen et preparatlarının, TMAB ve koliform bakterisi sayıları bakımından önemli farklılıklar gözlemlendi (p<0.001). Et numunelerinin TMAB sayısı 2.79-3.65 log10 kob/g arasında tespit edildi. En yüksek TMAB sayısı, gerdan kısmında (3.65 log10 kob/g), en düsük deger ise kontrnuarda (2.79 log10 kob/g) belirlendi. Numunelerin koliform bakteri sayısı 2.72-3.22 log10 kob/g arasında degistigi, en yüksek degerin nuar da (3.22 log10 kob/g), en düsük degerin ise yumurtada (2.72 log10 kob/g) oldugu gözlemlendi. Et parçalama ünitesinden temin edilen et preparatlarının E. coli ve koagulaz pozitif Staphylococcus sayıları bakımından önemli farklılıklar tespit edildi (p<0.001). Et numunelerinin E. coli sayısı, 2.47-2.67 log10 kob/g arasında belirlendi. Bu mikroorganizmanın bazı et preparatlarında (antrekot, kontrnuar, yumurta, gerdan, nuar, bonfile) belirlenebilir seviyenin altında oldugu gözlemlendi. Numunelerin koagulaz pozitif Staphylococcus sayısının, 2.52-2.83 log10 kob/g arasında degistigi, yumurta, kontrfile, tranç ve bonfile de ise ) belirlenebilir seviyenin altında oldugu tespit edildi. Marketlerden temin edilen numunelerde TMAB, koliform, E. coli ve koagulaz pozitif Staphylococcus sayıları bakımdan istatistiki farklılıklar tespit edildi (p<0.001). Marketlerden temin edilen et preparatlarının TMAB sayısı 3.49-4.18 log10 kob/g arasında belirlendi. Numunelerin koliform bakteri sayısının 2.80-3.71 log10 kob/g arasında belirlendi. En düsük deger bonfile de (2.80 log10 kob/g), en yüksek degerin ise sokum da (3.71 log10 kob/g) gözlemlendi. Marketlerden temin edilen et preparatlarından yumurta, nuar, kontrnuar ve bonfilede E. coli; yumurta, kontrfile ve bonfilede ise koagulaz pozitif Staphylococcus üremesi tespit edilebilir düzeyin altında bulundu. Diger et preparatların E. coli sayısının 1.76-2.60; koagulaz pozitif Staphylococcus sayısının ise 2.60-3.17 log10 kob/g arasında degisim gösterdigi belirledi. ncelenen kıyma numunelerinin TMAB sayısı 5.22; koliform bakteri sayısı 4.51; E. coli sayısı 2.61 ve koagulaz pozitif Staphylococcus 3.42 log10 kob/g olarak tespit edildi. Kıyma numunelerinde incelenen mikrorganizmalar açısından mevsimsel olarak istatistiki açıdan önemli düzeyde farklılıklar belirlendi (p<0.001) ve incelenen mikroorganizmaların en yüksek degerine yaz mevsiminde ulastıgı gözlemlendi. Sonuç olarak Antalya'da tüketime sunulan kırmızı et preparatları ve kıymaların mikrobiyal florasının diger bazı illere (Örn, Kayseri, Van) göre daha yüksek kalitede oldugu, ancak Türk Gıda Kodeksi ve Gıda Mevzuatında belirtilen sınır degerlere yakın oldugu gözlemlendi. Bu bakımdan bütün dünyada uygulama alanı bulan HACCP (Hazard Analysis and Critical Control Points) sisteminin etkinliginin gıda isyerlerinde daha etkin ve uygulanabilir kılınması gerektigi kanaatine varıldı.
In this study, ribeye, silverside, knuckle, striploin, topside, rump, neck eyeround, and tenderloin preparations were taken separately during twelve months both from markets and meat processing unit of company which is active in Antalya. Twenty four samples were taken from each of the meat pieces giving a total of 432 samples used in this research. In addition 60 minced beef samples were taken from different markets. All samples are examined and evaluated statistically from the point of view of the total aerobic mesofilic bacteria, coliform bacteria , E. coli and coagulase positive Staphylococcus. Significant differences were observed in the total mesofilic aerobic mesofilic bacteria and coliform bacteria number of meat preparations which were taken from the meat processing unit (p<0.001). The total mesofilic bacteria count of meat samples was determined as 2.79-3.65 log10 cfu/g. The highest value was found in the neck (3.65 log 10 cfu/g) and the lowest value was in the silverside (2.79 log10cfu/g). It was observed that the coliform bacteria counts of samples were in the range from 2.72 log10 cfu/g to 3.22 log10 cfu/g and the highest value belonged to the eyeround and the lowest value was in the knuckle. Significant differences were determined between the E. coli and coagulase positive Staphylococcus counts of meat preparations taken from meat processing unit (p<0.001). E. coli number of meat preparations was determined between 2.47-2.67 log 10 cfu/ g. It was observed that these microorganisms did under detectable limits in some meat preparations (ribeye, silverside, knuckle, neck, eyeround and tenderloin). It was found that sample?s coagulase positive Staphylococcus count ranged from 2.52 log 10 cfu/g to 2.83 log10 cfu/g and also observed that this microorganism was under detectable limits in the knuckle, striploin, topside and tenderloin. Statistical differences were determined in the number of the total aerobic mesofilic microorganisms, coliform bacteria, E. coli and coagulase positive Staphylococcus numbers of samples which were taken from markets (P<0.001). The total aerobic mesofilic bacteria number of meat preparations taken from markets determined to be between 3.49-4.18 log 10 cfu/g. Coliform bacteria numbers of the samples were determined to be between 2.80- 3.71 log 10 cfu/g. The lowest value was detected in the tenderloin as 2.80 log 10 cfu/g and the highest value in the rump as 3.71 log 10 cfu/g E. coli, growth was under detectable limits in the knuckle, eyeround, silverside and tenderloin taken from the markets and coagulase positive Staphylococcus growth was under detectable limits in samples of knuckle, eyeround, silverside and tenderloin. E. coli number of other meat samples determined as 1.76 -2.60 log 10 cfu/g and coagulase positive Staphylococcus number determined between 2.60-3.17 log 10 cfu/g. The total aerobic mesofilic bacteria counts of the minced beef samples were determined as 5.22; coliform bacteria number as 4.51; E. coli number as 2.61 and cougulase positive Staphylococcus number as 3.42 log 10 cfu/g. In minced beef samples, differencies were determined seasonally in terms of the investigated microorganism (P<0.001) and it was observed that investigated microorganism reached highest value in summer. As a result, microbial flora of red meat samples and minced beef which are present to the consumer in Antalya have a higher quality when compared with some other cities (e.g. Kayseri, Van). However the microbial value observed was near to the limit value which is detected in Food Codex and Food Regulation. In this respect the HACCP system which has an application field in all over the world have a necessity to be more effective and applicable in food plants.
