Bu araştırma, Konya ve çevresinde üretilen, deri, bez ve bidon olmak üzere üç farklı tip ambalaj içerisinde olgunlaştırılan, market ve pazar şeklinde iki farklı satış merkezinde tüketime sunulan tulum peynirlerinin kimyasal ve mikrobiyolojik özelliklerini belirleyerek; ambalaj şeklinin ve satışa sunuldukları alanların kalite niteliklerine etkisini saptamak amacıyla gerçekleştirildi. Kimyasal yönden analizi yapılan numune gruplarının, rutubet, kuru maddede yağ, kuru maddede tuz ve kül miktarları ile laktik asit cinsinden asitlik ve pH dereceleri sırasıyla %38.50-49.77, %27.26-49.17, %5.60-8.42, %3.63-5.58, %0.72-1.07 ve 5.02-5.38 değerleri arasında tespit edilmiştir. Satış noktaları dikkate alındığında tulum peynirlerinin, rutubet (P<0.001), kuru maddede tuz (P<0.05) ve kül (P<0.01) miktarlarında, gruplar arasında çıkan fark önemli bulunmuş olup, pazarda tüketime sunulan numunelerin rutubet, kuru maddede tuz ve kül miktarları yönünden, markette satılanlara göre daha yüksek olduğu belirlenmiştir. Ambalaj şekilleri dikkate alındığında tulum peynirlerinin rutubet (P<0.01), kuru maddede yağ (P<0.01) ve kül (P<0.05) miktarlarında gruplar arasında çıkan fark önemli bulunmuş olup, bez ambalajlarda tüketime sunulan numunelerin, rutubet ve kül miktarı bakımından en yüksek, bununla birlikte kuru maddede yağ miktarı için ise en düşük değere sahip olduğu görülmüştür. Mikrobiyolojik yönden analizi yapılan numune gruplarının, toplam aerob mezofilik, toplam aerob psikrofilik, maya-küf, Lactobacillus, koliform, E. coli ve Staphylococcus mikroorganizma sayıları sırasıyla 7.43-8.05 log10 kob/g, 3.22-4.57 log10 kob/g, 4.49-6.54 log10 kob/g, 7.24-7.71 log10 kob/g, 2.50-4.35 log10 kob/g, 2.45-4.04 log10 kob/g ve 4.51- 5.84 log10 kob/g değerleri arasında tespit edilmiştir. Bununla birlikte örneklerin hiç birinde Salmonella spp. tespit edilmemiştir. Satış noktaları dikkate alındığında tulum peynirlerinin, maya-küf sayısı yönünden, gruplar arasında çıkan fark önemli bulunmuş olup (P<0.05), pazarda tüketime sunulan numunelerin maya-küf sayısının, markette satılanlara göre daha yüksek olduğu belirlenmiştir. Ambalaj şekilleri dikkate alındığında tulum peynirlerinin toplam aerob mezofilik mikroorganizma sayıları yönünden, gruplar arasında çıkan fark önemli bulunmuş olup (P<0.05), bez ambalajlarda tüketime sunulan numunelerin en düşük toplam aerob mezofilik mikroorganizma sayısına sahip olduğu ve diğer ambalaj tiplerinden farklı olduğu belirlenmiştir. S. aureus yönünden identifikasyon yapılan toplam 291 koloninin, %81.44'ü S. aureus, %3.09'u atipik S. aureus, %2.41'i S.epidermidis, %2.41'i Micrococcus, %6.87'si Streptococcaceae (örn., Streptococci, Enterococci) familyası üyesi, %3.09'u koagulaz pozitif mannitol negatif diğer Staphylococcus türleri ve %0.69'u ise koagulaz negatif, glikoz ve mannitol pozitif diğer Staphylococcus türleri olarak identifiye edilmiştir. Satış noktaları dikkate alındığında S. aureus'un oransal olarak dağılımı %78.79 ile %83.65 arasında belirlenmiş ve en yüksek oran pazar grubunda tespit edilmiştir. Ambalaj materyalleri dikkate alındığında ise S. aureus'un oransal olarak dağılımı %73.61 ile %85.37 arasında tespit edilmiş ve en yüksek oran deri tulum, en düşük oran ise bidon grubunda tespit edilmiştir. Analizi yapılan toplam 100 numunenin 12'sinde (%12) enterotoksin saptanmıştır. Buna göre enterotoksin tip A, 2 numunede, enterotoksin tip B, 1 numunede, enterotoksin tip C, 9 numunede, enterotoksin tip D, 1 numunede ve enterotoksin tip E ise 1 numunede tespit edilmiştir. Ambalaj materyalleri dikkate alındığında enterotoksin oranı en yüksek bidon grubunda (% 28), en düşük ise bez grubunda (% 5.8) tespit edilmiştir. Satış noktaları dikkate alındığında en yüksek oran pazar grubunda (%12.9) belirlenmiştir. Sonuç olarak incelenen tulum peyniri örneklerinin, Salmonella spp. tespit edilmemesine rağmen, E.coli, koliform bakterisi, psikrofilik mikroorganizmalar ve mayaküf yönünden, sayılarının standardın üstünde bir değerde tespit edilmesi bununla birlikte 12 numunede enterotoksin saptanması ve diğer numunelerde de enterotoksin oluşturabilecek düzeyde Staphylococcus mikroorganizmalarının bulunmasından dolayı, halk sağlığı açısından potansiyel bir risk taşıdığı ayrıca tulum peyniri örneklerinin tüketime sunulduğu noktalar ve ambalaj materyalleri değişkenlerinin de ürünün kalitesini hem kimyasal hem de mikrobiyolojik yönden etkilediği kanaatine varıldı.
In this study, to determine chemical and microbiological properties of tulum cheese which produced in Konya and in the vicinity of Konya, ripened in three different packages as skin bag, cloth bag and plastic drum, submitted to consumption in two different sales outlets as store and market and effects of package types and sales outlets to quality characteristics were aimed. Moisture, fat in dry matter, salt in dry matter and ash values, acidity in terms of lactic acid and pH values of sample groups which analysed chemically were detected as %38.50- 49.77, %27.26-49.17, %5.60-8.42, %3.63-5.58, %0.72-1.07 and 5.02-5.38, respectively. The difference between groups for moisture (P<0.001), salt in dry matter (P<0.005) and ash values (P<0.01) of tulum cheese taking into account for sales outlets were detected statistically significant and moisture, salt in dry matter and ash values of samples which submitted to the consumption in markets were determined as higher than samples which sold in stores. The difference between groups for moisture (P<0.001), fat in dry matter (P<0.001) and ash values (P<0.05) of tulum cheese taking into account for packages materials were detected statistically significant and moisture and ash values were determined in the highest values and fat in dry matter values were determined in the lowest values in samples which submitted to consumption in cloth packages. The number of total aerobic mesophilic, total aerobic psicrophilic, yeast-mould, Lactobacillus, Coliform, E. coli and Staphylococcus microorganisms in samples which analised microbiologically were detected as 7.43-8.05 log10 cfu/g, 3.22-4.57 log10 cfu/g, 4.49-6.54 log10 cfu/g, 7.24-7.71 log10 cfu/g, 2.50-4.35 log10 cfu/g, 2.45-4.04 log10 cfu/g and 4.51-5.84 log10 cfu/g, respectively. In addition to these, no Salmonella spp. were detected in each samples. The difference between groups for the number of yeast-mould of tulum cheese taking into account for sales outlets was detected statistically significant (P<0.05) and the number of yeast-mould in samples which submitted to consumption in market was determined higher than samples which sold in stores. The difference between groups for numbers of total aerobic mesophilic microorganisms of tulum cheese taking into account for different packages was determined statistically significant (P<0.05) and total aerobic mesophilic microorganisms number was detected as the lowest in samples which submitted to consumption in cloth packages. Totally 291 colonies were identified for S. aureus and %81.44 of them were identified as S. aureus, %3.09 of them were identified as atypical S. aureus, %2.41 of them were identified as S. epidermidis, %2.41 of them were identified as Micrococcus, %6.87 of them were identified as members of Streptococcaceae (e.g., Streptococci, Enterococci) family, %3.09 of them were identified as coagulase positive mannitol negative other Staphylococcus species and %0.69 of them were identified as coagulase negative, glucose and mannitol positive other Staphylococcus species. Average distribution of totally 291 colonies identified for S. aureus taking into account for sales outlets were determined between %78.79 and %83.65 and the highest average was detected in market group. Average distribution of S. aureus taking into account for packaging materials was determined between %73.61 and %85.37 and the lowest average was detected in plastic drum group. Enterotoxine was detected in 12 samples of totally analysed 100 samples (%12). According to these, enterotoxine type A, enterotoxine type B, enterotoxine type C, enterotoxine type D and enterotoxine type E were detected in 2, 1, 9, 1 and 1 samples, respectively. Taking into account for packaging materials the highest enterotoxine average was determined in plastic drum group (% 28) and the lowest enterotoxine average was determined in cloth group (% 5.8). Taking into account for sales outlets the highest enterotoxine average was determined in market group (%12.9). As a result, in spite of no detection of Salmonella spp. in investigated tulum cheese samples, in terms of E.coli, Coliform bacterium, psicrophilic microorganisms and yeast and mould numbers detected at a higher level than the standards and due to the detection of enterotoxine in 12 samples and the presence of Staphylococcus microorganisms at a level that they had ability to produce enterotoxine, it was believed that they had a potential risk for public health and places where tulum cheese samples submitted for consumption and package materials effected the product quality in terms of chemical and microbiological properties.
