Bu çalısmada, Selçuk Üniversitesi Veteriner Fakültesi ve Konya Veteriner Kontrol ve Arastırma Enstitüsü'ne, 2007-2008 kuzulama döneminde, Konya il ve ilçe merkezleri ile köylerinde bulunan 79 ayrı koyun sürüsünden yavru atma sikayeti ile getirilen 86 adet atık koyun fetusundan izole edilen 24 adet B. melitensis susu ile Konya Veteriner Kontrol ve Arastırma Enstitüsü'nce, 2006- 2007 kuzulama döneminde Konya bölgesine ait atık koyun fetuslarından izole edilmis 12 adet B. melitensis susunun konvansiyonel testleri ve biyotiplendirmesi yapıldı. Sahadan izole edilen B. melitensis suslarının RAPD yöntemi ile DNA parmak izleri belirlenerek genotiplendirmeleri ve birbirlerine olan genetik yakınlıkları tespit edilerek yaygın olarak bulunan genotip belirlendi. B. melitensis Rev-1 ası susu da RAPD yöntemi ile değerlendirilip saha suslarının bant modelleri ile karsılastırıldı. Ayrıca, atık materyallerinden izole edilen B. melitensis saha suslarının, B. melitensis Rev-1 ası susundan köken alıp almadığı PZR-RFLP yöntemi ile arastırıldı. Atık görülen koyun sürülerinden kan serumları toplanarak RBPT, mSAT ve iELISA testleri ile değerlendirilerek enfeksiyonun yaygınlığı tespit edildi. Biyotiplendirme sonucunda, 36 tane B. melitensis izolatının 3 tanesinin biyotip 1, 32 tanesinin biyotip 3 ve bir tanesinin B. melitensis Rev-1 ası susu olduğu belirlendi. Saha izolatı B. melitensis Rev-1 ası susu moleküler olarakda PZR-RFLP yöntemiyle teyit edildi. 9, 24, 25, 27, 28, 30, 31, 32 ve 34 numaralı izolatların, penisilin veya tiyonin ve bazik fuksine duyarlılık göstermeleri nedeniyle atipik oldukları saptandı. Otuz altı adet izolatın B. melitensis olduğu, tür spesifik AMOSPZR metodu ile de doğrulandı. RAPD-PZR metodu kullanılarak, saha susları ile B. melitensis Rev-1 ası susunun DNA parmakizleri çıkarıldı ve UPGMA ile NJ dendogramları olusturuldu. Her iki dendogramda da susların 14 alt gruba ayrıldığı ve aynı dağılımları sergiledikleri görüldü. Konya Bölgesi'nde atığa sebep olan 36 adet B. melitensis izolatının aynı kaynaktan köken almadığı saptandı. Aynı alt grupta yer alan 10 adet izolatın en yaygın görülen genotip olduğu gözlendi. Farklı alt gruplarda yer alan atipik susların farklı klonlardan köken aldığı belirlendi. Referans suslar ile 36 adet B. melitensis saha izolatları 109 arasında ki genetik benzerlik değerleri, % 50.0'den % 99.9'a kadar genis bir aralıkta dağılım gösterdi. B. melitensis Rev-1 ası susunun, RAPD-PZR ile identifiye edilemeyeceği belirlendi. B. melitensis izole edilen 24 koyun sürüsünden elde edilen toplam 588 adet koyun kan serumu örneği, RBPT, mSAT ve iELISA testleri ile değerlendirildi ve sırasıyla % 41.1, % 37.2 ve % 43.2 pozitiflik belirlendi. Sonuç olarak, Konya Bölgesi'nde atıklara sebep olan B. melitensis suslarının RAPD-PZR yöntemi ile farklılık gösterdiği, en çok izole edilen ve tiplendirilen biyotipin biotip 3 olduğu ve ası susu B. melitensis Rev-1'in ilk kez bir atık fetüsten izole edildiği ortaya konuldu. RAPD-PZR yöntemin uygulama ve yorumlamasının basit olduğu, Brusellozis'in epidemiyolojisinde kullanılmasının fayda sağlayacağı kanaatine varıldı.
In this study, by a total of 86 aborted sheep fetuses brought from different sheep herds (n=79) in lambing season 2007-2008 from Konya and its rural areas to Selcuk University Veterinary Faculty and Konya Veterinary Control and Research Institute numbers of 12 and of 24 Brucella melitensis (B.melitensis) were isolated and identified by the Faculty and the Research Institute, respectively. The strains isolated isolated from aborted sheep fetuses by Konya Veterinary Contol and Research Institute in lambing season 2006-2007 were identified by conventional tests and biotyping procedures. DNA fingerprints, genetic relationships and common genotypes of field isolated of B. melitensis strains were determined by RAPD method. B. melitensis Rev-1 vaccine strain was checked by RAPD method and fingerprint obtained was compared with those of field isolates. In addition, B. melitensis strains isolated from field were evaluated by RFLP-PZR to determine origins of them for belonging to the vaccine strain Rev-1. Serum samples were collected from infected sheep herds (n=24) and evaluated by RBPT, mSAT and iELISA tests in order to determine nature of spread of infection. Of the 36 B. melitensis isolates, 3, 32 and 1 were identified as biotype 1, biotype 3 and B. melitensis Rev-1 vaccine strain, respectively. Field isolated B. melitensis Rev-1 vaccine strain was confirmed by RFLP-PZR. Isolates of 9, 24, 25, 27, 28, 30, 31, 32 and 34 were detected for being atypical since they were susceptible of penicillin or thionin and basic fuschin. A total of 36 B. melitensis isolates were confirmed by species specific AMOS-PZR. DNA fingerprints of field isolates and B. melitensis Rev-1 vaccine strain were obtained by RAPD-PZR method and UPGMA and NJ dendograms were created. Strains were separated similarly 14 subgroups in both dendograms. 36 B. melitensis isolates from Konya region were detected that they had not from the same source. A number of 10 isolates located in the same subgroup were observed the most common genotypes. Atypical strains that located in different subgroups have different clonal origin. Levels of genetic similarity between reference strains and field isolates were 50,0-99,9%. That between reference vaccine strain B. melitensis Rev-1 and field isolated B. melitensis Rev-1 was 111 88,9%. It was also detected that B. melitensis Rev-1 vaccine strain cannot be identified by RAPD-PZR alone, while isolates located the same main group with B. melitensis Rev-1 can be suspected for being the vaccine strain. A total of 588 serum samples collected from B. melitensis isolation positive herds (n 24) were examined by RBPT, mSAT and iELISA tests. Of the 588 serum samples, 41,1%, 37,2% and 43,2% were found to be positive by RBPT, mSAT and iELISA, respectively. Consequently, B. melitensis strains were isolated in Konya region were found to be different by RAPD-PZR. B. melitensis biotype 3 was found the most common biotype. B. melitensis Rev-1 vaccine strain were isolated from aborted fetus in Türkiye for the first time. RAPD-PZR method was found for being an ease to use and interpretation. This can be used for studies on epidemiology of Brucellosis.
