Wnt genlerinin bir çok türde reprodüktif olayların düzenlenmesinde önemli rollerinin olduğu bilinmektedir. Fakat Wnt genleri kısrak endometriumunda henüz karakterize edilmemiştir. Bu çalışmanın amacı siklik kısraklarda siklusun ovulasyon günü (0. gün) geç diöstrus ve luteolizis sonrası östrus ile birlikte gebeliğin 15., 18. ve 22. günlerinde Wnt sinyal iletim sistemine ait genlerin ekspresyon seviyelerinin mRNA düzeyinde belirlenmesidir. Bu amaç için yukarıda bahsedilen her bir gün için 4'er adet olacak şekilde reprodüktif yönden sağlıklı kısraklardan biyopsi örnekleri toplanmıştır. Siklusun günleri kan progesteron düzeyi ve ultrasonografi ile tespit edilmiştir. Ayrıca gebeliklerin teşhisinde de utrasonografiden yararlanılmıştır. Toplanan biyopsi örneklerinden total RNA izole edilerek RT reaksiyonu ile cDNA'ya çevrilmiştir. Kısrak endometriumda PZR ile 17 Wnt geni içinden Wnt2, Wnt2b, Wnt4, Wnt5a, Wnt5b Wnt7a, Wnt8a, Wnt9b, Wnt10b, Wnt11, Wnt16, Wnt sinyal iletim sistemi antagonistlerinden SFRP1, SFRP2, SFRP5, Dkk1, Dkk2, Wif-1, hücre yüzey reseptörü FZD6 ve LRP6, intrasitoplazmik kompanentlerden DVL-2, GSK3-beta, Beta-catenin, nükleer faktör Lef-1 ve Wnt genlerinin aktive ettiği Myc ile MSX2 genlerinin endometriumda siklik ve gebe günlerin tamamında ekspresyon tespit edildi. Real-time PZR ile bu genlerin siklik ve gebe günlerdeki ekspresyon farklılıkları araştırılmıştır. Normalize edilmiş data mixed model ANOVA kullanılarak analiz edildi ve gruplar arası farkların belirlenmesi için least significant difference test kullanıldı. Siklus günleri içerisinde Wnt2b, Wnt5a, Wnt9b, Wnt10b, Wnt11, Wnt16, Dkk1, Wif-1 ve Myc genlerinin geç diöstrus döneminde miktarları önemli ölçüde artmıştır (P<0,05). Wnt7a ve SFRP1 geç diöstrus döneminde baskılanmıştır (P<0,05). Wnt5b ekspresyonu geç diöstus ve luteolizis sonrası östrusta ovulasyona göre azalmıştır (P<0,05). Wnt4 ve Lef-1 ise sadece luteolizis sonrası östrusta artmıştır (P<0,05). Erken gebelikte ovulasyon günü ile kıyaslandığından Wnt2, Wnt5b, Wnt7a ve SFRP1 gebelik günleri içerisinde baskılanmıştır (P<0,05). Non-canonical Wnt5a, Wnt11 ile antagonist Wif-1'in ekspresyonu gebeliğin 15. ve 18. günlerinde artmıştır (P<0,05). Wnt2b ise sadece gebeliğin 15. gününde artmıştır (P<0,05). Dkk1'in gebelik süresince ekspresyonu artmıştır (P<0,05). Wnt16 ise erken gebelik boyunca artış eğilimindedir (P<0,09). Siklusun geç diöstrus dönemi ile gebeliğin 15. günü kıyaslandığında, Wnt8a (P<0,05) ve Myc (P<0,08 ) gebeliğin 15. gününde baskılanmış, Wnt4'ün ekspresyonu artmıştır (P<0,06). Luteolizis sonrası östrus ile 18. gün gebelik kıyaslandığında Wnt7a, SFRP1, Lef -1 baskılanmış, Dkk1, Wif-1 ve MSX2 genleri ise aynı dönemde ekspresyonları artış göstermiştir (P<0,05). Aynı karşılaştırma günlerinde Wnt5a ve Wnt16 gebeliğin 18. gününde artış eğilimindedir (P<0,09). Sonuç olarak; Wnt gen ailesi ve bunlara ait sinyal iletim sistemi kompanentleri ilk defa kısrak endometriumunda karakterize edilmiştir. Elde edilen verilere bakıldığında östrus esnasında bu genlerden bazılarının ekspresyonunun maternal hormonların etkisi ile değiştiği söylenebilir. Benzer bir şekilde erken gebeliğin bu genlerin ekspresyonunu düzenlediğinin gösterilmesi, Wnt genlerinin daha henüz uterus içerisine implante olmayan embriyonun büyüyüp gelişmesi için erken gebelik boyunca endometriumda proliferasyon ve sekresyon fonksiyonlarının düzenlenmesinde rol oynayabileceğini göstermektedir
Wnt genes have important roles in reproductive events of many species. To our knowledge, there is no data concerning the Wnt family genes expression in equine endometrium. The objective was to characterize Wnt genes in equine endometrium in different stages of estrous cycle and early pregnancy. Expression of Wnt genes and their steady state mRNA levels were evaluated from endometrial biopsy samples of reproductively sound mares on day of ovulation (d0, n=4), late diestrus (LD, n=4) and after luteolysis in estrus phase (AL, n=4) of the estrous cycle. The stages of estrous cycle were confirmed by plasma progesterone concentrations measured daily and ultrasound examinations. Also, on days 15 (P15), 18 (P18) and 22 (P22) of early pregnancy, biopsy samples were collected (n=4 for each day). Pregnancies were confirmed by ultrasonography. Total RNA was extracted and converted to cDNA by using Reverse Transcriptase reaction. PCR amplifications showed that out of 17 Wnt genes, Wnt2, Wnt2b, Wnt4, Wnt5a, Wnt5b, Wnt7a, Wnt8a, Wnt9b, Wnt10b, Wnt11 and Wnt16 were present in the endometrium in all stages of estrous cycle and pregnancy. Also, Wnt antagonists SFRP1, SFRP2, SFRP5, Dkk1, Dkk2, Wif-1, cell membrane receptors FZD6 and LRP6, cytoplasmic DVL-2, GSK3-beta, Beta-catenin, nuclear transcription factor Lef-1 and Wnt target genes Myc and MSX2 expressions were observed in both cyclic and early pregnant endometrium. Relative quantifications were made by Real time-PCR. A mixed model of ANOVA was fitted on the normalized data and least significant difference test was employed to determine significantly different groups. Wnt2b, Wnt5a, Wnt9b, Wnt10b, Wnt11, Wnt16, Dkk1, Wif-1 and Myc were upregulated in LD compared to d0 and AL (P<0.05). Wnt7a and SFRP1 expressions were downregulated in LD (P<0.05). Wnt5b expression was attenuated in both LD and AL compared to d0 (P<0.05). Wnt4 and Lef-1 expressions were only upregulated in AL compared to d0 and LD (P<0.05). In early pregnancy, Wnt2, Wnt5b, Wnt7a ve SFRP1 expression were downregulated compared to d0 (P<0.05). Non-canonical Wnt5a, Wnt11 and antagonist Wif-1 were upregulated on days P15 and P18 (P<0.05). Wnt2b only increased on day P15 (P<0.05). Pregnancy increased the amount of Dkk1 (P<0.05). During the early pregnancy, increase in Wnt16 expression was slightly significant (P<0,09). Compared to LD, Wnt8a (P<0.05) and Myc (P<0.08) decreased on P15, while Wnt4 was upregulated (P<0.06). When AL and P18 were compared, Wnt7a, SFRP1, Lef-1 expressions were downregulated while Dkk1, Wif-1 ve MSX2 were upregulated (P<0.05). Also, in the same comparison days, Wnt5a and Wnt16 tended to be upregulated on day P18 (P<0.09). In conclusion, Wnt genes, their antagonists and their associated signal transduction components were, the first time, characterized in equine endometrium. Some of Wnt genes were regulated by the maternal hormones during the estrous cycle. Similarly, these genes also seem to be regulated during early pregnancy, therefore it can be suggested that regulation of some Wnt genes in endometrium may be mediating the endometrial proliferative and secretory functions to support the growth and nourishment of unattached embryo in uterus during early pregnancy.
