Organik bir insektisit olan fipronil, pestisitlerin fenil pirazoller veya fiproller olarak bilinen daha yeni ve küçük bir grubuna dahildir. Fipronil genis spektrumlu bir insektisit olup, zirai alanda, çevre sağlığında ve veteriner hekimlikte pek çok zararlıya karsı etkilidir ve tüm dünyada kullanımı artmaktadır. Bu çalışmanın birinci amacı saf ve ticari fipronilin tavuk embriyolarının gelişimi üzerindeki olası embriyotoksik ve teratojenik etkilerinin belirlenmesiydi. Bu amaçla asetonda çözdürülmüş saf fipronil ve steril bidistile suda sulandırılmış ticari fipronil solüsyonları kuluçka başlangıcında döllü tavuk yumurtalarının hava kamarasına enjekte edildi. Yumurtalar kuluçkanın 15. gününde açılarak grupların ölü ve anormal embriyo sayıları, malformasyon tipleri, canlı ve nispî embriyo ağırlıkları ile
embriyoların tepe-kıç boyları (Crown-Rump Length, CRL) belirlendi. Regresyon analizi yöntemiyle saf fipronilin LD50 (Letal Doz %50) değeri 161 μg/yumurta, ticari fipronilin D50 degeri 383 μg/yumurta olarak tayin edildi. Saf ve ticari fipronil grupları ile kontrol grubu arasında anormal embriyo oranı bakımından farklar istatistiksel öneme sahip değildi. Bununla birlikte saf ve ticari fipronil gruplarında canlı ve rölatif embriyo ağırlıkları ile embriyoların CRL değerlerinin kontrol grubuna kıyasla önemli düzeyde azalma gösterdiği belirlendi. Bu sonuçlar, hem saf hem de ticari fipronilin metabolize olmamış formunun tavuk embriyoları üzerinde önemli embriyotoksik etkiye sahip olduğunu, bununla birlikte teratojenik etkiye sahip olmadığını göstermektedir. Çalışmanın ikinci aşamasında LD50 ve subletal dozlarda (161 μg/yumurta, 80.50 μg/yumurta ve 40.25 μg/yumurta) saf fipronil kuluçka başlangıcında ve kuluçkanın 8. gününde hava kamarası yoluyla döllü tavuk yumurtalarına enjekte edildi. Pozitif kontrol olarak aflatoksin B1 ve siklofosfamid kullanıldı. Kuluçkanın farklı günlerinde ve kuluçka sonunda fipronilin embriyotoksik, teratojenik ve genotoksik etkileri yanı sıra immün sistem üzerindeki olası zararlı etkileri değerlendirildi. Kuluçkanın 8. gününde enjekte edilen fipronilin kuluçka öncesi yapılan enjeksiyona kıyasla daha toksik etki gösterdiği belirlendi. Ayrıca Alizarin
Red-S ile yapılan boyamalarda, özellikle kuluçkanın 8. gününde enjeksiyon yapılan
tüm fipronil gruplarındaki embriyolarda kuluçkanın 11. gününde iskelet sistemi
gelişiminde gecikme gözlendi. Bu sonuçlar, tavuk embriyolarında bu günlerde karaciğerin fonksiyonel olgunlaşması ve geniş bir metabolik aktivite sahası
bulunmasıyla bağlantılı olarak fipronilin metabolitlerinin doğal formundan daha
toksik olduğuna işaret etmektedir. Kuluçka başlangıcında fipronil verilen yumurtalardan elde edilen 10 günlük civcivlerin periferal kanındaki akyuvar sayısı, lenfosit ve Alfa Naftil Asetat Esteraz-pozitif (ANAE(+)) lenfosit oranları kontrol gruplarına kıyasla önemli düzeyde azalma gösterdi. Bu veriler metabolize olmamış fipronilin hücresel immünite üzerinde zararlı etkiye sahip olabileceğini göstermektedir. Mikronukleus (MN) testi sonuçları ise bu çalışmada uygulanan dozlardaki fipronilin ve metabolitlerinin genotoksik etkiye sahip olmadığına işaret etmektedir.
Fipronil, an organic insecticide, is a member of relatively new and small class of pesticides, the phenyl pyrazoles or fiproles group of chemicals. Fipronil is a highly effective, broad-spectrum insecticide that effectively controls a wide range of crop, public hygiene and veterinary pests and its use worlwide is increasing. The first aim of this study was to determine the possible embryotoxic and teratogenic effects of both pure and commercial formulations of fipronil on the development of chicken embryos. For this purpose, solutions of pure fipronil dissolved in acetone and commercial fipronil diluted with distilled water (20 µl/egg) were injected into the air space of fertilized hen?s eggs at the beginning of the incubation. The eggs were opened at 15th day of the incubation and the following parameters of each group were examined: dead and abnormal embryo numbers, malformation types, both live and relative embryo weights and crown-rump lengths (CRL). Regression analysis estimated the LD50 (the 50% lethal dose) for pure fipronil and commercial fipronil to be 161 µg/egg and 383 µg/egg, respectively. The differences in abnormal embryo rates were not statistically significant between the control and the fipronil groups. However, the live and relative embryo weights and embryo CRL values of the fipronil groups were significantly lower than that of the control group. Based on the results, it was concluded that non-metabolized forms of both pure and commercial fipronil have significant embryotoxic effects but no teratogenic effects on chicken embryos. In the second stage of this study, LD50 and sublethal doses of pure fipronil (161 µg/egg, 80.50 µg/egg and 40.25 µg/egg) were injected into the air space of fertilized hen?s eggs at the beginning and 8th day of the incubation. Aflatoxin B1 and cyclophosphamide were also used as positive controls. Embryotoxic, teratogenic and genotoxic effects of fipronil were evaluated on different days of the incubation and after hatching in addition to its possible deleterious effects on the immune system. Fipronil which was injected at 8th day of the incubation had more toxic effect than that of the pre-incubation injection. Besides, the results of Alizarin Red-S staining demonstrated that all of the fipronil doses injected, especially, at 8th day of the incubation caused retardation in the development of the embriyonic skeletal system at 11th day of the incubation. These results indicated that metabolites of fipronil were more toxic than its native form to chicken embryo, related to the functional maturation of liver and a wide range of metabolic activities occurring at these days. In the peripheral blood samples of 10 days-old chickens obtained from the eggs which were treated with fipronil at the beginning of the incubation, leucocyte counts, lymphocyte and Alpha Naphtyl Acetate Esterase-positive (ANAE(+)) lymphocyte proportions were significantly decreased when compared to the control groups. These data revealed that non-metabolized fipronil might have harmful effect on the cellular immunity. The results of micronucleus (MN) assays pointed out that fipronil and its metabolites had no genotoxic effect at doses tested.
