Nörotrofinler, özellikle periferik sinir sisteminde hücrelerin gelişim, farklılaşma, canlılık ve metabolizma gibi hayati işlevleri etkileyen büyüme faktörleridir. Polipeptid yapıda, suda eriyebilen bu faktörler nöron kas ve bağ dokusu gibi hedef dokulardan salgılanır ve retrograt aksonal taşınılma gövdeye taşınabilir. Etkilerini iki tip reseptöre bağlanarak (yüksek affiniteli tirozin kinaz A, B, C ve düşük affinite gösteren p75 reseptörleri) gösterirler. Çalışmada bu reseptörelerin arka kök gangliyondaki expresyon düzeyleri araştırıldı. Her iki cinsten Balb-C fareler den stereomikroskop altında çıkarılan arka kök gangliyonları %4 paraformaldehid tespitinden sonra 10-50 mikrometre alınan kriyo kesitler anti TrkA, B,C ve P75 primer antikorları ve fluorokrom bağlı 488, 564. 594 sekonder antikorları ile işaretlendi. Fluoresan ve konfokal mikroskopta incelenerek dijital olarak görüntülendi. En kuvvetli immün yanıt gösteren NT reseptörü Trk A ve daha sonra Trk B iken; en az immün yanıt gösteren Trk C ve P75 idi. Gruplar arasında da ortalama olarak tüm reseptörlere en fazla yanıt gösteren erişkin erkek grubu ve daha sonra erişkin dişi grubu iken; en az yanıt gösteren genç dişi grubu idi. Sonuç olarak; sinir fizyolojisinde önemi yadsınamaz olan nörotrofinlere ait reseptörlerin arka kök gangliyonu olarak adlandırılan primer duysal nöron topluluklarında değişik oran ve tarzda ifade edildiği immünhistokimyasal olarak gösterildi.
Neurotrophins are trophic factors which affect vital functions as development, differentiation, viability and metabolism of nervous system cells. These soluble and polypeptide form factors secreted from neuron and other target tissues as muscle and connective tissues and transported retrogradely from akson to the soma. Their effects were acted via two types of receptor (high affinity tyrosine kinase A, B, C and a low affinity receptor p75). This study was aimed to determine the expression of these receptors. For this purpose, L4 and L5 spinal ganglions were taken bilaterally from mice. After fixation (paraformaldehyde 4%) and sucrose (30%) processing, 5-30 ?m thickness sections were taken by frozen section and then labeled anti Trk A, B,C and P75 primary and fluorescent conjugated secondary antibodies. Tissue samples were examined with confocal and fluorescent microscopes and imagines were captured digitally. In the confocal microscopic examination of sections: the strongest immunreactivity were in the Trk A and then Trk B while the weakest were of Trk Cand p75 respectivelly. Between the groups of all receptors, showing the highest response were adult male and then adult female groups while while showing the least response was the young female group. As a result, the undeniable importance of neurotrophins in neural physiology of the receptor of primary sensory neurons in dorsal root ganglion, expression of these were shown immunohistochemically in different proportions and manner
