Konya iline bağlı 14 ilçe (Akşehir, Beyşehir, Bozkır, Cihanbeyli, Çumra, Ereğli, Halkapınar, Ilgın, Kadınhanı, Karapınar, Karatay, Meram, Selçuklu, Seydişehir), Karaman'ın il merkezi ve Ayrancı ilçesi olmak üzere 16 farklı bölgede, farklı yaş gruplarına ait (< 6 ay, 6 ay-1 yaş arası, 1-3 yaş arası ve > 3 yaş) 250 keçi ve 985 koyun olmak üzere toplam 1235 hayvanın her birinin kulak ucundan sürme kan frotileri yapılmıştır. Hayvanların vena jugularisinden de antikoagulantlı ve antikoagulantsız yaklaşık 10'ar ml kan alınmış ve serumlar çıkarılmıştır. Kan frotileri Giemsa boyama yöntemi ile boyandıktan sonra ışık mikroskobu ile 100X objektif altında etken varlığı yönünden incelenmiştir. Serumlardan Anaplasma ovis (A. ovis) tespitinde Competitive Enzyme Linked Immunosorbent Assay (cELISA), Anaplasma phagocytophilum (A. phagocytophilum) tespitinde ise Indirekt Floresan Antikor (IFA) yöntemlerinden yararlanılmıştır. Mikroskobik muayenede koyunların %9,8'i (97/985), keçilerin %10,4'ü (26/250); cELISA testi ile koyunların %84,5'i (484/573), keçilerin ise %53,4'ü (132/247) A. ovis yönünden pozitif bulunmuştur. Antikoagulantlı kanların santrifüj edilerek elde edilen buffy coat tabakasından hazırlanan ince yayma frotilerin hiçbirinde mikroskobik inceleme sonucu A. phagocytophilum tespit edilememiştir. 832 hayvana (585 koyun, 247 keçi) ait kan serumu, A. phagocytophilum antijen preparatları kullanılarak IFA testi ile anti- A. phagocytophilum antikor varlığı yönünden araştırılmıştır. IFA testi sonucunda koyunların 83'ü (%14,2), keçilerin 94'ü (%38,1) olmak üzere toplam 177 (%21,3) küçük ruminant A. phagocytophilum enfeksiyonu yönünden seropozitif olarak tespit edilmiştir.
A total of 1235 small ruminants (250 goats and 985 sheep) belonging to the different age groups (<6 months, between 6 months and 1 year old, 1-3 years old and > 3 years) in 16 different areas including 14 districts (Aksehir, Beysehir, Bozkir, Cihanbeyli, Cumra, Eregli, Halkapinar, İlgin, Kadinhani, Karapinar, Karatay, Meram, Selcuk, Seydisehir) from Konya province and two distrits (city centre and Ayrancı) from Karaman province were examined for Anaplasma infections. Giemsa stained peripheral blood smears prepared from eartip of all animals were screened under the 100X light microscope for the presence of Anaplasma ovis organisms. Serum samples were tested to determine specific antibodies against Anaplasma ovis (A. ovis) and Anaplasma phagocytophilum (A. phagocytohilum) by Competitive Enzyme Linked Immunosorbent Assay (cELISA test) and Indirect Fluorescent Antibody test (IFA), respectively. The results of microscopic examination showed that 9.8% (97/985) of the sheep and 10.4% (26/250) of the goats were positive for Anaplasma ovis organisms. The cELISA results revealed that 84.5% (484/573) of the sheep and 53.4% (132/247) of the goats were sero-positive for A. ovis-specific antibodies. None of the thin smears prepared from buffy coat layer of anticoagulated blood were positive for the presence of A. phagocytophilum organisms by microscopy. IFA results showed that totally 177 (21.3%) animals including 83 sheep (14.2%) and 94 goats (38.1%) had specific antibodies against A. phagocytophilum.
