Metallotioninler, hücre içi çinko dengesinin düzenlenmesi ve fizyolojik etkilerine aracılık etmelerinin yanı sıra güçlü antioksidan etkileriyle de dikkat çeken proteinlerdir. Bu çalışmanın amacı da sıçanlarda testis, böbrek ve kalp iskemi-reperfüzyon hasarında çinko uygulamasının metallotioin düzeylerini nasıl etkilediğinin araştırılmasıdır. Çalışma üç ayrı projede kullanılan 94 adet Wistar-Albino cinsi erişkin erkek sıçanlardan elde edilen testis, böbrek ve kalp dokuları üzerinde gerçekleştirildi. Çalışma protokolü Selçuk Üniversitesi Veteriner Fakültesi "Canlı Hayvan Kullanılmayacak Araştırmalar İçin Etik Kurul Raporu" etik kurulu tarafından onaylandı. Deney Hayvanları: 1.Testis çalışma grupları: Kontrol (T-Kont.), Sham (T-Sh.), İskemi/Reperfüzyon (T-İ/R.) ve çinko uygulanan İskemi /Reperfüzyon (T/Zn-İ/R). 2.Böbrek çalışma grupları: Kontrol (B-Kont.), Sham (B-Sh), İskemi/Reperfüzyon (B-İ/R) ve çinko uygulanan İskemi /Reperfüzyon (B/Zn-İ/R). 3.Kalp çalışma grupları: Kontrol (K-Kont), Kalp İskemi/Reperfüzyon (K-İ/R) ve çinko uygulanan İskemi /Reperfüzyon (K/Zn-İ/R) olmak üzere toplam 11 gruba ayrıldı. Sham ile testis, böbrek ve kalp Iskemi-reperfüzyon uygulamaları genel anestezi altında yapıldı. Çinko İ/R gruplarına ise 21 gün süreyle sıçan başına 5 mg/kg gün periton içi (i.p.) çinko sülfat verildi. Uygulamaların bitiminde tüm hayvanlar dekapite edilerek testis, böbrek ve kalp dokuları alındı. Bahsedeilen dokularda Rat metallotionin antikoru ile immünohistokimyasal boyama prosedürleri gerçekleştirildi. Boyanan preperatlar fotoğraflandı ve metallotionin boyanmış hücreler ışık mikroskobunda sayılarak yüzdeleri hesaplandı. Testis ve böbrek doku örneklerinde çinko desteği metallotionin boyanmış hücre sayılarını diğer gruplara oranla önemli şekilde arttırdı (p<0.05). Kalp doku örneklerinde ise grupların metallotionin düzeyleri arasında amlamlı bir farklılık elde edilmedi. Çalışmanın sonucunda elde edilen bulgular çinko uygulamasının özellikle böbrek ve testis iskemi reperfüzyonunda metallotionin sentezini uyararak antioksidan aktiviteyi artırıcı etkiye sahip olabileceğini göstermektedir.
Metallothioneins, as well as mediating physiological effects and intracellular zinc balance and and attracting attention are proteins with potent antioxidant. The aim of the present study was to search how zinc sulphate supplementation affects metallothionein levels testes, kidney and heart ischemia-reperfuison damage in rats. Study was performed on the testes, kidney and heart tissue which obtain from 3 different Project, total 94 Wistar-Albino type adults male rats. Experimental protocols were approved by Selcuk University, Veterinary School, Ethics Committee. Animals groups for testes: Control (T-Cont.), Sham (T-Sh.), ischemia /reperfusion (T-I/R.) and Zinc supplemented ischemia /reperfusion (T/Zn-I/R), for kidney: Control (K-Cont.), Sham (K-Sh), ischemia/reperfusion (K-I/R), Zinc supplemented ischemia/reperfusion (K/Zn-I/R), for heart: Control (H-Cont), Heart ischemia/reperfusion (H-I/R) and Zinc supplemented ischemia/reperfusion (H/Zn-I/R) which totally were 11 groups. Sham groups testes, kidney and heart ischemia/reperfusion were performed under general anesthesia. For Zinc I/R groups, zinc supplemented 5 mg/kg/day for 3 week by intraperitoneally. After supplementations all were decapited and testes, heart and kidney tissues taken. In mentioned tissue immunohistochemical staining procedures were conducted by rat metallothionein antibody. The stained preparations were photographed and metallothionein stained cells under a light microscope by counting and calculated the percentage of stained cells. In the testes and kidney tissues, zinc supplementation has increased metallothionein stained cell number compared to the other groups (p<0.05). However, in the heart tissue, metallothionein levels were not different among the groups. The results of present study show that zinc supplementation induces MT synthesis especially in kidney and testes ischemia-reperfusion and may have antioxidant activity by stimulating the synthesis metallothionein.
