Doku spesifik genler gelişim ve metabolizma da önemli roller oynamaktadır. Günümüzde, gen bankasında farklı dokularla ilgili 3.363.628 GEO Profil ve 397.988 mikroarray verisi bulunmaktadır. Bilim adamları için gen bankasında bu kadar çok faydalı veri bulunmasına rağmen, yeni doku spesifik genlerin ortaya çıkarılması için bu verileri analiz edip kullanmaya yönelik bir strateji izlenmesiyle ilgili eksiklikler bulunmaktadır. Bu çalışma, mikroarray ve mikroarraylerle ilişkili GEO Profillerin kullanımı ve analiziyle doku spesifik genlerin ortaya çıkarılmasına yönelik yeni bir metot ortaya koymaktadır. Bu çalışmada, adipoz doku bu metodun doğrulanmasında hedef doku olarak seçilmiştir. İlk olarak, insan ve fare mikroarray verileri karşılaştırmalı olarak analiz edilmiştir. Mikroarray verilerini desteklemek için, adipoz dokuyla ilgili GEO Profilleri gen bankasından seçilmiştir. Daha sonra, adipoz dokuyla ilgili microarray ve GEO Profil verileri eş zamanlı olarak değerlendirilmiştir. Sonuç olarak bu metot, Chordin-like 1 (Chrdl1) olarak isimlendirilen yeni bir adipoz spesifik geni hipotez etmiştir. Hipotezin testi için sığır dokularında RT-PCR yapılmıştır. RT-PCR sonuçlarına göre, Chrdl1 diğer dokulara göre adipoz dokuda yüksek derecede ekspresyon göstermiştir (P<0.05). Bu sonuç, kurulan hipotezi desteklemiş ve bu çalışmada kullanılan metodun yeni doku spesifik genlerin belirlenmesinde faydalı olabileceğini göstermiştir.
Tissue specific genes play important roles in development and metabolism. Currently, Gen Bank has 3.363.628 GEO Profiles and 397.988 microarray data related to various tissues. Although huge amounts of useful data are available to scientists in Gen Bank, there is a lack of a follow-up strategy to analysis and use these data to identify new tissue specific genes. This study suggest a unique new method to using and analyzing microarray and microarray related GEO Profiles to identify tissue specific genes. In this study, adipose tissue sellected as a target tissue in order to validate the method. First, the human and mouse microarray data were analyzed comparatively. To support the microarray data, adipose tissue related GEO Profiles were selected from Gen Bank. Subsequently, adipose tissue related microarray and GEO Profiles data were evaluated simultaneously. Finally, this method hypothesized a novel adipose specific gene called Chordin-like 1 (Chrdl1). In order to test the hypothesis, RT-PCR was performed for the bovine tissue distribution. According to the RT-PCR results, Chrdl1 was highly expressed adipose tissue versus other tissues (P<0.05). This result was support the hypothesis and demonstrated that a new method used in this study was useful in identification of new tissue specific genes.
