Bu çalıĢmada yavaĢ embriyo dondurma yöntemi ve bazı kriyoprotektanların dondurulup çözdürülmüĢ Saanen keçisi embriyolarının canlılığı üzerine etkilerinin belirlenmesi amaçlanmıĢtır. Projenin hayvan materyalini 15 adet Saanen ırkı diĢi keçi ve 3 adet teke oluĢturdu. Hayvanlardan yeterli embriyo toplanabilmesi için iki sezon uygulama yapıldı. Keçilere 12 gün süreyle 20 mg Fluorogeston asetat emdirilmiĢ intravaginal sünger (Chronogest CR, Ġntervet, Holland) yerleĢtirildi. Sünger uygulandıktan sonraki 9. gün itibariyle 3 gün 12 saat arayla azalan dozlarda FSH (2,5; 1,5; 1 cc) enjekte edildi. Sünger 12. gün çıkarıldı ve 24 saat sonra doğal aĢım yaptırıldı. Ġlk aĢımdan sonraki 7. gün laparoskopik uterus yıkaması yapılarak embriyolar elde edildi. Toplanan embriyolar değerlendirilerek kaliteli olanlar etilen glikol, gliserol ve DMSO kullanılarak yavaĢ dondurma yöntemiyle donduruldu. Bu amaçla. Her kriyoprotektan grubu için ortalama 30 embriyo donduruldu. Embriyolar sıvı azot içerisinde çödürülüp değerlendirilene kadar muhafaza edildi. Embriyolar 37 º C‟de su banyosunda çözdürüldü. Çözdürme sonrasında embriyolar 38,5 ºC‟de % 5 CO2‟de inkübasyona bırakıldı. Embriyo geliĢimleri 24, 48 ve 72. saatlerde değerlendirilidi. Elde edilen verilerin istatistiki analizi % t testi kullanılarak yapıldı. Birinci ve ikinci sezonda bütün hayvanlarda östrus görüldü. Süperovulasyon cevapları ( 4≥ Cl) sırasıyla % 93,3 ve % 84,6, embriyo toplama oranı ise % 61,4, % 72,3 olarak belirlendi. Hayvanların % 26,7 ve % 15,4‟inde erken luteal regresyon oranı tespit edildi. Transfer edilebilir embriyo oranı birinci sezonda % 58, ikinci sezonda ise % 61,6 olarak belirlendi. Çözdürme sonrasında canlılık oranlarında, bütün gruplarda saatlerin önemli derecede etkili olduğu ve 72. saate ulaĢmada isatistiki farkın oluĢtuğu görüldü (p<0,05). Gruplar arasında etilen glikolle dondurulan blastosislerin DMSO ile dondurulanlardan daha iyi olduğu (p<0,05) ancak grup içerisinde değerlendirildiğinde morulaların canlılıklarında 24-72. saatler arasında önemli farklılıkların (p<0,05) olduğu görüldü. Saanen keçisi embriyolarının yavaĢ yöntemle dondurulması sırasında kriyoprotektan olarak en iyi sonucun etilen glikolle alındığı ve 72. saate ulaĢma da etilen glikolün özellikle DMSO olmak üzere her iki kriyoprotaktandan daha baĢarılı olduğu görüldü. Elde edilen bilgiler ıĢığında etilen glikolün keçi embriyolarının yavaĢ yöntemle dondurulmasında baĢarıyla kullanılabileceği kanaati oluştu.
In this study, aim was to determine effects of slow embryo freezing method and some cryoprotectants on the viability of the Saanen goats embryos Animal material of this project was 15 Saanen goats and 3 bucks. In order to collect enough embryos, animals were used in two consecutive breeding seasons. Fifteen goats were synchronized with intravaginal sponges containing 20 mg of Fluorogeston acetate (Chronogest CR, Ġntervet, Holland) for 12 days. Decreasing doses of FSH (2,5; 1,5; 1 cc) were injected at 12 hour intervals on the last 3 days of sponge treatment. Twentfour hours after sponge removal, goats were allowed to mate. Laparoscopic uterine washing was performed and embryos were collected on the seventh day after the first mating. Collected embryos were assessed and embryos with good quality were frozen with a slow freezing method using different cryoprotectants. For this purpose ethylene glycol, glycerol and DMSO were used. An average of 30 embryos were frozen with each cryoprotectant. Embryos were stored in liquid nitrogen and then were thawed in 37 ° C water bath.After thawing, the embryos were incubated at 38.5 º C and 5% CO2. Viability controls at 24, 48 and 72 hours were performed. Statistical analysis of data was performed using the % t test. Estrus was observed in all animals both in the first and the second season. Superovulation response (4≥ Cl) was found to be 93.3% and 84.6% for two seasons, respectively. Recovery rate is 61.8% and 72.3%. Early luteal regression was detected in 26.7% and 15.4% of animals respectively in two seasons. The rate of transferable embryos was 58% and 61.6%, respectively. In the viability rates after thawing, hours was significantly effective and in achieving to 72 hours there was statistical difference in all groups (p <0.05). Betweeen groups, survival rates of blastacytes with frozen ethylene glycol was better than that with frozen DMSO (p <0.05). However, within the groups the viability of the morulas significant differences were observed between 24-72 hours (p <0.05). During slow freezing method, the best results were obtained by ethylene glycol. In the achieving to 72 hours, ethylene glycol was found to be more successful than other cryoprotectants. Consequently, ethylene glycol can be used successfully during slow freezing method for goat embryos.
