TGFβ, organizmanın tüm dokularının gelişiminde, homeostazisinde ve onarımınında önemli rolleri olan gelişimi çok yönlü kontrol eden önemli bir polipeptitdir. TGFβ meme kanseri hücrelerinin invazyon ve proliferasyonunu düzenleyen güçlü bir düzenleyicidir. Vitamin D,kalsiyum ve fosfor homeostazisi klasik etkisinin yanında anti-proliferatif, anti-invazif, anti-neoplastik, pro-apoptotik etkileri olan pleotropik bir hormondur. Epidemiyolojik ve deneysel in vivo ve vitro çalışmalarda kanser üzerine etkinliği gösterilmiştir. Bizim çalışmamızda MCF-7 meme kanseri hücrelerinin proliferasyonu üzerindeTGFβ2 ve D vitamininin etkisini ölçmeyi amaçladık. TGFβ2 ve D vitamininin proliferasyon üzerine etkisini gerçek zamanlı hücre elektronik algılama sistemiyle (xCELLigence) ortaya koymayı amaçladık. Hücre sayısındaki değişiklikler, mikro elektrot içeren özel hücre kültürü kuyucuklarında, deney süresince her 15 dk'da bir sürekli olarak izlendi. Proteaz çalışmaları için uPA aktivite ve PAI-1 protein seviyesi ölçmek amaçlanmıştır. TGFβ2 ve vitamin D uygulanan MCF-7 hücrelerindeki proliferasyon inhibisyonu doz ve zaman bağımlı olarak gözlendi. TGFβ2 ve D vitamini için belirlenen IC50 dozu uPA aktivitesi ve PAI-1 düzeyi analizleri için hesaplanmıştır. TGFβ2 ve D vitamini her iki IC50 dozu ile muamele edilen lizatlarda uPA aktivitesi ve PAI-1 düzeyi hesaplandı. uPA aktivitesi ve PAI-1 protein ekspresyonu hem TGFβ2 hem de D Vitamini ile engellenmiştir. TGFβ2 uPA aktivitesi ve PAI-1 protein ekspresyonunu sadece inhibe ederken, D vitamini ise ikili bir etki göstemiştir. Sonuç olarak, TGFβ2 ve vitamin D'nin meme kanseri hücresi proliferasyonunu doz ve zaman bağımlı olarak inhibe ettiği ve bunu uPA aktivitesini ve PAI-1 düzeyini azaltarak gösterdiği gözlenmiştir. Bu değerlendirmeyle birlikte, TGFβ2 ve vitamin D'nin hücre üzerine etki mekanizmasıyla ilgili daha ileri çalışmalar ve hiperkalsemik olmayan analoglarının geliştirilmesi ile eksiklikler giderilebilir.
TGFβ is a polypeptide which has an important role of the development homeostasis and repair of all the tissues. TGFβ is a potent regulators regulate breast cancer cell invasion and proliferation. 1,25(OH)2D3, is a pleiotropic hormone with anti-proliferative, pro-apoptotic and pro-differentiative effects on numerous cell types, which suggest anti-cancer activity in addition to its classical regulatory action on calcium and phosphate metabolism. Epidemiological and experimental in vitro and in vivo data support a cancer preventive role of 1,25(OH)2D3. In our study, we aimed to measure the effect of TGFβ2 and vitamin D effect on the MCF-7 breast cancer cells proliferation . The cell proliferation studies were done by on line and real-time cell electronic sensing system (xCELLigence). Changes in the number of cells, the particular cell culture in wells containing micro-electrodes, a duration of the experiment was continuously monitored every 15 min. For protease studies we aimed to measure uPA activity and its inhibitör PAI-1 protein level. Dose and time dependent inhibition of proliferation were observed in TGFβ2 and vitamin D treated MCF-7 cells. Inhibitor concentration of 50% doses were calculated from the software for both TGFβ2 and vitamin D. The cells lysates treated by IC50 doses for both TGFβ2 and vitamin D were taken for uPA activity and PAI-1 protein. uPA activity and PAI-1 protein expression were inhibited by both TGFβ2 and vitamin D. Vitamin D showed dual effect on the system while TGFβ2 showed onlyu inhibiton of both PAI-1 protein level and uPA activity. In conclusion, proliferation of breast cancer cell was inhibited by TGFβ2 and vitamin D via dose-time dependent decreasing uPA activity and PAI-1 level were observed. Further studies of the mechanisms effect of TGFβ2 and vitamin D on cell proliferation and the development of analogues with non-hypercalcemic deficiencies rectified.
