Bu çalışmanın amacı sıçanlarda ligatürle oluşturulan deneysel periodontitiste sodyum selenit ve/veya alfa-tokoferol uygulamalarının alveoler kemik kaybı, dişeti kollajen lif sayısı, uyarılabilir nitrik oksit sentaz (iNOS)+ ve CD95+ hücre sayıları ve serum sitokin konsantrasyonları üzerine olan etkilerini incelemektir. Dört haftalık çalışmada, sıçanların sol alt molar dişinin dişeti kenarına 3/0 ipek sütur bağlandı. Daha sonra, her grupta on adet olacak şekilde toplam kırk Sprague Dawley sıçan dört gruba ayrıldı: Grup A (K): kontrol grubu, intraperitoniyal (IP) serum fizyolojik enjeksiyonu, Grup B (Se): sodyum selenit grubu, (0.2 mg/kg/gün, IP), Grup C (?T): alfa-tokoferol grubu (40 mg/kg/gün, IP), Grup D (Se + ?T): kombine grup, alfa-tokoferol ve sodyum selenit'in kombine kullanıldığı grup (40 mg/kg/gün ve 0.2 mg/kg/gün IP). Dokular histometrik ve immünohistokimyasal inceleme için %10'luk formaldehit solüsyonu ile fikse edildi. Alveoler kemik kaybı (AKK) Clemex Vision Lite 3.5 görüntü analiz yöntemiyle değerlendirildi. Birleşim epitelinin altındaki bağdokusunda iNOS, CD95 pozitif hücre ve kollajen lif sayıları görüntü analiz sistemi kullanılarak sayıldı. Serum interlökin (IL)-1ß, IL-6 ve IL-4 konsantrasyonları ELISA kitleriyle ölçüldü. İstatistiksel analiz için Kruskal-Wallis Testi ve Bonferroni düzeltmeli Mann Whitney U testi yapıldı Çalışmanın sonunda Se+?T'nin birlikte kullanıldığı grup AKK'yı kontrol grubuna göre önemli derecede baskıladı (p<0,05). Serum sitokin konsantrasyonları açısından gruplar arasında fark yoktu (p>0,05). Grup D'deki kollajen lif miktarı kontrol grubundan daha fazlaydı, ancak fark önemli değildi (p>0,05). Grup B'deki iNOS+ sayısı Grup A'dakinden düşüktü (p<0,05). Grup C'deki histometrik, immünohistokimyasal ve biyokimyasal parameterelerin hiçbiri Grup A'ya göre önemli bir fark göstremedi (p>0,05). Kombine olarak Se+?T uygulanması iNOS+ ve CD95+ hücre sayısını kontrollere kıyasla azalttı, ancak fark anlamlı bulunmadı (p>0,05). The aim of this study was to evaluate the effects of sodium selenite and/or alpha-tocopherol administration on alveolar bone loss, number of gingival collagen fibers, inducible nitric oxide synthase (iNOS)+ and CD95+ cells and serum cytokine concentrations in rats with ligature induced periodontitis. In this four-week study, 3/0 silk sutures were placed at the gingival margin of the lower right first molars in a mandibular quadrant. Forty male Sprague Dawley rats were then divided into four groups of ten rats each: Group A (C): a control group, saline intraperitoneal injection (IP), Group B (Se): sodium selenite group (0.2 mg/kg/day, IP), Group C (?T): alpha-tocopherol group (40 mg/kg/day IP), and Group D (Se+?T): combined group, alpha-tocopherol used in combination with sodium selenite (0.2 mg/kg/day and 40 mg/kg/day, IP). The tissues were fixed using neutral buffered 10% formaldehyde solution for histometrical and immunohistochemical examination. Alveolar bone loss (ABL) (?m) was measured by Clemex Vision Lite 3.5 vision analysis system. The number of iNOS, CD95 positive cells and collagen fibers in subepithelial connective tissue beneath the sulcular epithelium were counted by using the same image analysis system. The concentrations of serum interleukin (IL)-1ß, IL-6 and IL-4 levels were measured by using ELISA kits. Kruskal-Wallis test and Bonferroni adjusted Mann Whitney U tests were used for the statistical analysis. At the end of the study, the combination of Se and ?T suppressed ABL compared to saline group (p<0,05). There was no statistically significant differences in serum cytokine concentrations among the study groups (p>0,05). Higher levels of collagen fiber counts were found in Group D compared to controls, but differences were not significant (p>0,05). The number of iNOS+ cells in Group B was at lower levels than Group A (p<0,05). Histometrical, immunohistochemical and biochemical parameters in Group C were not significantly different than Group A (p>0,05). The combination of Se and ?T administration decreased the number of iNOS+ and CD95+ cells compared to the controls, but there was no significant difference (p>0,05). We concluded that sodium selenite suppressed iNOS-induced gingival inflammation in rat models. Within the limits of this study, it can be suggested that sodium selenite and/or alpha-tocopherol administration may be useful to prevent periodontal tissue destruction.
