Bu çalışmanın amacı, deneysel diyabet oluşturulan sıçanlarda, silimarin takviyesinin kognitif fonksiyonlar üzerine etkisini incelemekti. Çalışmada 38 adet 400-450 gr ağırlığında Wistar cinsi erkek sıçan kullanıldı. Sıçanlar rastgele seçilerek kontrol, silimarin, diyabet ve diyabet+silimarin gruplarına ayrıldı. Diyabet ve diyabet+silimarin gruplarına tek doz streptozotosin (50 mg/kg) uygulandı. Uygulamadan 7 gün sonra kan glikoz konsantrasyonları ölçülerek diyabet doğrulandı ve silimarin ve diyabet+silimarin gruplarına 35 gün süreyle 200 mg/kg silimarin oral yolla verildi. Çalışmanın 30-35. günlerinde sıçanlara, Açık Alan (OF) Testi, Yükseltilmiş Artı Labirent (EPM) Testi ve Moris Su Labirenti (MWM) Testi uygulandı. Sıçanlardan 35. günün sonunda anestezi altında kan ve beyin numuneleri alındı. Alınan numunelerde beyin kaynaklı nörotrofik faktör (BDNF) ve Histondeasetilaz (HDAC) seviyeleri ölçüldü. Sıçanların kan glikoz seviyeleri diyabet gruplarında kontrol ve silimarin gruplarından daha yüksekti. OF testinde defekasyon sayısı, diyabet+silimarin ve silimarin gruplarında diyabet grubuna göre daha düşüktü (P<0,05). EPM testinde kapalı kollara giriş sayısı diyabet gruplarında kontrol grubuna göre yüksekti ve açık kollarda geçirilen süre diyabet grubunda kontrol grubuna göre düşüktü (P<0,05). MWM testinde platformdan geçiş sayısı ve platform alanında geçirilen süre diyabet grubunda kontrol grubundan düşüktü (P<0,05). Plazma ve beyin dokusu BDNF seviyeleri diyabetli grupta diğer tüm gruplardan düşüktü (P<0,05), HDAC seviyeleri gruplar arasında farklı değildi (P>0,05). Sonuç olarak bu çalışmadan elde ettiğimiz bulgular, silimarin takviyesinin diyabetli sıçanlarda BDNF seviyelerini artırarak kognitif fonksiyonları iyileştirebileceğini ileri sürmektedir.
The aim of this study was to examine the effects of silymarin supplementation on cognitive
functions in experimentally-induced diabetic rats. In this study 38 male Wistar rats, weighing between 400-450 g were used. Rats were randomly divided into the control, silymarin, diabetes and diabetes+silymarin groups. Diabetes and diabetes+silymarin groups were injected single dose streptozotocin (50 mg/kg). After seven days of the injection, diabetes was confirmed by measuring blood glucose concentration and silymarin and Diabetes+silymarin groups were given 200 mg/kg silymarin via orally for 35 days. On the 30th-35th days of the study, Open Field (OF) Test, Elevated Plus Maze (EPM) Test and Moris Water Maze (MWM) Test were performed. At the end of the 35th day, brain and blood samples were taken from the rats under anesthesia. Brain-derived neurotrophic factor (BDNF) and histone deacetylase (HDAC) levels were measured in these samples. Blood glucose levels were higher in diabetic groups than control and silymarin groups. In the
OF test, the number of defecation was lower in the silymarin and diabetes+silymarin groups than diabetic group (P<0.05). In the EPM test, the number entrance to the closed arms were higher and time spent in open arms was lower in the diabetic group than the control group (P<0.05). In MWM test, the number of platform crossing and time spent in the platform area was lower in the diabetic group than control group (P<0.05). In the diabetic group, plasma and brain tissue BDNF levels were lower than the other groups (P<0.05), in the HDAC levels there was no difference among the groups (P>0.05). In conclusion, the results obtained from this study suggest that, silymarin supplementation could improve cognitive functions in diabetic rats by increasing the BDNF levels.
