Bu çalışma; agronomik özellikleri açısından ümitvar bulunan 25 adet kabak genotipinin kuraklık
stresine tolerans seviyesini belirlemek amacıyla yapılmıştır. Seçilen genotiplere ait bitkiler tam kontrollü
iklim odası koşullarında hidroponik ortamda Hoagland besin çözeltisi içeren saksılarda kültüre alınmıştır.
Usulüne uygun olarak yetiştirilen fidelerde gerçek yaprak oluştuğunda besin çözeltisine -0,42 MPa
ozmotik potansiyele denk gelen %6 oranında Polietilen Glikol (PEG 6000) eklenerek kuraklık stresi
oluşturulmuştur. Uygulama sonrasında kontrol ve kuraklık uygulanmış kabak genotiplerinde stres
belirtileri görülmeye başladığı anda kök ve yapraklarında yaş ağırlık (g), kuru ağırlık (g), sürgün ve kök
boyları (cm), yaprak bağıl su içerikleri (%) ve iyon sızıntısı incelenmiştir. Araştırma sonucunda çerezlik
kabak genotiplerinin büyüme ve gelişmesi kontrol grubu ile mukayese edilerek değişimleri incelenmiş ve
kuraklık stresine karşı genotiplerin gösterdikleri tolerans düzeyleri ortaya konulmuştur. PEG 6000 teşvikli
kuraklık stresine bağlı olarak çerezlik kabak genotiplerinde yaş ve kuru ağırlık ile sürgün ve kök boyunun
azaldığı ve bu azalmanın kontrol uygulamasına göre en fazla B-25, A32, A-18, A5, A1, A33, C-24
genotiplerinde olduğu belirlenmiştir. Araştırmada kullanılan çerezlik kabak genotipleri içerisinde B-33 ve
A-7 genotiplerinde kurak koşullarda büyüme ve gelişiminde azalmanın diğer genotiplere göre çok daha
az oranlarda olduğu tespit edilmiş ve bu genotipler kuraklığa toleransı en yüksek genotipler olarak
belirlenmiştir.
The experiment was conducted with the aim of determination of tolerance of 25 hopeful pumpkin
genotypes to drought stress for agronomical characteristics. Selected genotypes was placed in hydroponic
culture with Hoagland nutrient solution in full controlled under climate room conditions. When plants had
3-4 true leaves they were exposed to drought stress with -0,42 MPa osmotic potential by adding 6 %
polyethylene glycol (PEG 6000) to nutrient solution. Pumpkin control and drought treated plants after
application when stress symptoms begin to appear, some physiological parameters were investigated
including stem dry weight, root dry weight (g), leaf relative water content (%) and ion leakage. At the end
of study, pumpkin genotypes‟ changes in plant growth and development compared with control group
were investigated and genotypes‟ tolerance levels to drought stress were revealed. In PEG 6000 mediated
drought stress, in genotypes of pumpkin, fresh and dry weight and stem height of the body decreases and
maximum decrement was observed B-25, A32, A-18, A5, A1, A33, C-24 genotypes while compared with
control group. It was determined that there was no decreased in B-33 and A-7 genotypes compared with
control group and these genotypes were determined as the most tolerant ones to drought.
