Bir yapının rekonstrüksiyonunun en iyi yolu, o dokunun benzeri ile replasmanıdır. Kompozit doku allotransplantasyonu, deri, kemik, damar, sinir, kıkırdak gibi çeşitli dokular içeren anatomik yapıların transplantasyonu şeklinde tariflenmektedir. Allotransplantasyon sonrası akut ve kronik dönemde çeşitli biyokimyasal reaksiyonlar oluşmaktadır. Bu reaksiyonlar allogreft kaybına sebep olan en sık nedendir. Akut dönemde meydana gelen bazı değişikliklerin daha iyi araştırılmasına ihtiyaç vardır. Çalışmamızın amacı kompozit doku nakli alıcılarında, allogreft fonksiyonunun takibi, immün yükün oluşturduğu sistemik hasar ve akut rejeksiyon tanısında serum IL-6, IL-10, IL-18, TNF-α, Serum Amiloid A (SAA), C5a, CRP ve neutrophil gelatinase associated lipocalin (NGAL) seviyelerinin kullanılabilirliğini değerlendirmektir. Bu çalışmada, akut dönemde bazı serum değişkenleri incelenmiştir. İmmünsüpresyon uygulanmadan yapılan kompozit doku allotransplantasyonunun bu serum değişkenlerine etkileri araştırılmıştır. Transplantasyondan önce, transplantasyondan sonra birinci,üçüncü, ve beşinci günde kan örnekleri toplandı. Transplantasyondan beş gün sonra alıcı sıçanlar sakrifiye edildi. Kan değerleri karşılaştırılarak veriler toplandı. Bulgular istatistiksel analizlerle değerlendirildi. Biyokimyasal analizlerle elde edilen verilerin istatistiksel değerlendirmeleri sonucunda, çalışılan parametrelerden IL-6, IL-10, SAA ve CRP' nin akut dönemdeki değişimleri rejeksiyon reaksiyonlarını göstermede anlamlı bulunmuştur. Bununla birlikte daha spesifik ve sensitif parametrelerin tespitine ihtiyaç vardır.
The best way to reconstruct a structure is to replace it with its similar. Composite tissue allotransplantation is being described as transplantation of anatomical structures including skin, bone, vessel, nerve, cartilage. After allotransplantation, various biochemical reaxions occur in acute and chronic phases. These reaxions are the most common reason of allograft loss. Some changes that occur in acute phase need more research. The aim of our study is to evaluate the availibility of serum IL-6, IL-10, IL-18, TNF- α, serum amiloid A (SAA), C5a, CRP and neutrophil gelatinase associated lipocalin (NGAL) levels in the diagnosis of allograft function follow-up, systemic damage formed by immune load and acute rejection in composite tissue receivers. In this study, some serum variables were examined. The effects of composite tissue allotransplantation which performed without immunsuppressions on these serum variables were investigated. Blood samples were collected before transplantation, after first day, third day and fifth day of the transplantation. Receiver rats were sacrificed five day after the transpantation. Datas were collected bu comparing the blood values. Findings were evaluated with statistical analysis. As a result of evaluation of the datas' statistical analysis that collected from biochemical analysis, the acute phase changes of IL-6, IL-10, SAA and CRP was found significant for indicating the rejection reaxions among the studied parameters. However, it is need for detection of more specific and sensitive parameters.
