Bu tezde irinotekanın hemoglobin, globulin ve insan serum albumini gibi serum proteinlerine bağlanması çalışılmıştır. Bu amaçla epoksi modifiye süperparamagnetik demir oksit nanopartikülleri (GPTS-SPION) yüzeyi kullanılmıştır. Bu nanopartikül NH4OH ile Fe+2 ve Fe+3 tuzlarının birlikte çöktürülmesi ile sentezlenmiş ve sonra SPION yüzeyinde fonksiyonel epoksi grupları elde etmek için [3-2,3-epoksipropoksi)propil]trimetoksi silan (GPTS) ile modifiye edilmiştir. Sonuçlar sırasıyla 20mg GPTS modifiye SPION yüzeyine insan serum albumini (HSA) için 44,1, globulin (Glb) için 21,2 ve hemoglobin (Hem) 32,6 µg bağlandığı bulunmuştur. İrinotekanın serum proteinlerine bağlanma miktarlarının belirlenmesi amacıyla irinotekanın spektrofotometrik yöntemle uyarma ve yayılma dalga boyları belirlenmiştir. Buna göre uyarma dalga boyu 299 nm iken yayılma dalga boyu 448 nm bulunmuştur. 25 mg GPTS modifiye SPION yüzeyin serum protein-SPION yüzeylerine irinotekan bağlanma miktarları sırasıyla insan serum albumini (HSA) için 37,3, globulin (Glb) için 43,2 ve hemoglobin (Hem) için 40,8 µg bağlandığı bulunmuştur. Nanopartikül yüzeyine HSA, Glb ve Hem ile irinotekanın bağlanması infrared spektroskopisi ile doğrulanmıştır. Ayrıca yüzey karakterizasyonu için Taramalı Elektron Mikroskobu (SEM) analizi yapılmıştır. GPTS-SPION yüzeyinde serum proteini-irinotekan etkileşiminin termal davranışı termogravimetrik analiz (TGA) kullanılarak incelenmiştir. Termal parçalanmanın kinetik parametreleri Horowitz- Metzger metodu kullanılarak tayin edilmiştir.
The binding of irinotecan to serum proteins (hemoglobin, globulin, and human serum albumin) was studied on the surface of epoxide modified superparamagnetic iron oxide nanoparticles (GPTS-SPIONs), which were synthesized by the coprecipitation of ferrous and ferric salts with NH4OH and then modified with [3-(2,3-epoxypropoxy)propyl] trimethoxy silane (GPTS) to obtain functional epoxide groups on the SPIONs surface. Data showed that binding amount of human serum albumin (HSA), globulin(Glb) and hemoglobin (Hem) found to be as 44, 21.2 and 32.6 µg per 20 mg of GPTS modified SPIONs, respectively. The binding of irinotecan to serum protein was determined by using spectrofluorometer via excitation and emission wavelenght of the drug. The exicitation and emission wavelenghts of irinotecan was meassured as 299 nm and 448 nm, respectively. Data showed that binding amount of irinotecan to human serum albumin (HSA), globulin(Glb) and hemoglobin (Hem) found to be as 37.7, 43.2 and 40.8 µg per 20 mg of GPTS modified SPIONs, respectively. The binding of irinotecan to HSA-SPIONs, Hem-SPIONs and Glb-SPIONs was confirmed by using infrared spectroscopy scanning electron microscopy analysis was performed for surface characterization. The thermal behaviour of the serum protein-Ir interaction on GPTS-SPIONs was also studied by using thermo gravimetric analysis (TGA) technique and then the kinetic parametres for the thermal decomposition were determined using Horowitz-Metzger method.
