Araştırma, PTU enjeksiyonuyla hipotiroidizm oluşturulan ratlarda, L-tiroksin uygulamasının plazma vazopressin düzeyleri ve sıvı dengesinde oluşabilecek değişikliklerin ortaya konulabilmesi amacıyla planlandı. Çalışma için ağırlıkları 270-300gr arasında değişen 72 adet Wistar-albino türü erkek rat kullanıldı. Araştırmada 3 ana deney grubu oluşturulmuştur. (1. Grup: Kontrol grubu, 2. Grup: PTU-Kontrol grubu, 3. Grup: Deney (PTU+ L-tiroksin) grubu ). Ana gruplar her biri 6 hayvandan oluşan 4 alt gruptan meydana geldi. Bu alt gruplar; uyarım yapılmayan a grupları, izotonik uyarım yapılan b grupları, hipertonik uyarım yapılan c grupları ve hipovolemik uyarı yapılan d gruplarından oluşmaktadır. Deneylerin bitiminde ratlar dekapite edilerek kan örneklerinde hematokrit, ozmolalite, TSH, TT3, TT4 ve vazopressin tayini yapıldı. Ana grupların hematokrit değerleri kendi aralarında karşılaştırıldıklarında d (hipovolemik) grubunun hematokrit düzeyleri, diğerlerinden yüksek bulunmuştur (P<0.001). PTU grubunda hematokrit azalırken (P<0.001) PTU+tiroksin grubunda kontrole benzer olarak bulundu. Ozmolalite değerleri incelendiğinde; tüm ana grupların hipertonik uyarım yapılan gruplarında yüksek bulunmuştur (P<0.001). TSH değerleri PTU grubunda yüksek iken (P<0.001) PTU+tiroksin grubunda kontrolle benzer değerler bulunmuştur. Total T3 ve T4 değerleri PTU grubunda en düşük, tiroksin tedavisi yapılan grupta en yüksek olarak belirlendi (P<0.001). Vazopressin düzeyleri grup 1 ve 2'nin c ve d alt grupları diğer ikisinden de anlamlı yüksekti, d alt grupları ise hepsinden de yüksek bulunmuştur (P<0.001). Grup 3'de ise benzer şekilde c ve d alt grupları kendi aralarında anlamlı fark ifade etmezken, c ve d alt grupları diğer ikisine göre anlamlı olarak yüksekti (P<0.001). Alt grupların vazopressin değerleri incelendiğinde a, b ve c alt gruplarında PTU grubu en düşük, PTU+ tiroksin grubu ise en yüksek vazopressin düzeyleri görüldü. Hipovolemik uyarımlı gruplarda 3d her ikisinden de anlamlı olarak yüksek bulunmuştur (P<0.001). Bu çalışmanın sonuçları deneysel hipotiroidizmde bazal ve uyarılmış plazma vazopressin düzeylerinin azaldığını ancak hipotirodizmi takiben tiroksin tedavisinin bu azalmayı önlediğini göstermektedir. Bununla birlikte vazopressin salgısı için uyaranlar olan hipertonik ve hipovolemik uyaranlara karşı AVP cevabının tiroid fonksiyon bozukluğunda değişmediğini ortaya koymaktadır.
This study was performed to investigate the effects of L-thyroxine treatment on plasma vasopressin levels and possible changes in fluid balance in prophyltiouracil-induced hypothyroid rats. Total 72 Wistar-albino strain male rats weighing 270-300g were used for the study. Experiment animals were separated into three groups as follows (1. Group; General control group, 2. Group: PTU-Experiment group, 3. Group: PTU+ L-thyroxine group). Each main groups were divided to 4 subgroups including 6 rats for every group (a; unchallenged, b; isotonic stimulated, b;hypertonic stimulated, d;hypovolemic stimulated). At the end of the experiments all rats were decapited and plasma samples were analysed in terms of haematocrit, osmolility, TSH, TT3, TT4 and vasopressin. Haematocrit (Hct) levels were the highest in hypovolemic stimulated subgroups (P<0.001). In PTU+ Thyroxine group Hct levels were similar to control while in PTU group this parameter were reduced (P<0.001). Osmolality levels were higher in hypertonic stimulated groups (P<0.001). TSH leves in higher PTU group (P<0.001), but PTU+tyhyoxine group was similar to control group. Total T3 and T4 values were the lowest in PTU goup and the highest in tiroksin treatment group (P<0.001). Vasopressin levels in subgroups of c and d in groups 1 and 2 were higher than the other subgroups (a and b), and also d subgroup was the highest in all subgroups (P<0.001). Similar finding were found in group 3, c and d subgroups were higher than the other subgroups a and b (P<0.001). When the evaluated vasopressin levels in subgroups; vasopressin responses to unchallenged, isotonic and hypertonic stimulations were the lowest in PTU group (P<0.001). In hypovolemic subgroups, 3d subgroup has higher levels compared to other groups (P<0.001). The results of the present study show that basal and stimulated plasma vasopresin levels are reduced in hypothyroidism, however, thyroxine treatment following hypothyroidism prevents this reduction. On the other hand, the results exhibit that vasopressin response to hypertonic and hypovolemic stimulations are not changed in the thyroid dysfunction.
