Bu araştırmada, deneysel olarak propiltiyourasil (PTU) aracılığı ile hipotiroidizm ve L-tiroksin (L-T4) aracılığı ile hipertiroidizm oluşturulan ratlarda oksidatif stresin göstergelerinden tiyobarbitürik asit reaktif maddeleri (TBARS), nitrik oksit (NO) ve 8-hydroxy-2' -deoxyguanosine (8-OHdG) ile antioksidan savunma mekanizması bileşenlerinden süperoksit dismutaz (SOD), katalaz (CAT), glutatyon peroksidaz (GSH-Px), total glutatyon (GSH) ve total antioksidan kapasite (TAS) plazma düzeylerinde meydana gelen değişimlerin belirlenmesi amaçlandı. Araştırmada sağlıklı 70 adet, yaklaşık 300-350 g ağırlığında, 12 haftalık erkek Wistar Albino ırkı rattan yararlanıldı. Çalışmada 10 günlük adaptasyon periyodunun ardından rastgele seçilen 7 rattan alınan örneklerden elde edilen veriler, tüm gruplar için 0. gün değeri olarak kabul edildi. Daha sonra ratlar; kontrol, hipotiroid ve hipertiroid olmak üzere 3 gruba ayrıldı. Başlangıçta her bir grupta 7'şer adedi yedek olmak üzere 21 hayvan bulunmaktaydı. Bunların, 7'si 3. haftada, 7'si de 6. haftada kullanıldı. Hipotiroidizm ve hipertiroidizm sırasıyla PTU (6-n-propil-2-tiourasil; %0,05 w/v) ve L-T4 (L-tiroksin; %0,0012 w/v) içme sularına katılarak gerçekleştirildi. Araştırmada kullanılan ratların, canlı ağırlıkları ve yem tüketimleri kaydedildi. Uygulamaların başlatıldığı 0. gün ve araştırmanın 3. ve 6. haftalarında kontrol, hipotiroid ve hipertiroid gruplarındaki ratların kalplerinden, eter anestezisi altında kan alındı. Toplanan kanların, plazma ve serumları elde edildi. Serum örneklerinden, tiroid uyarıcı hormon (TSH), total triiyodotironin (TT3), serbest T3 (fT3), total tiroksin (TT4), serbest T4 (fT4) düzeyleri ve plazma örneklerinden TBARS, NO, 8-OHdG, SOD, CAT, GSH-Px, total GSH ve TAS düzeyleri belirlendi. Kontrol grubu 0. gün, 3. ve 6. hafta değerlerine göre, hipotiroid grubun 3. ve 6. haftalardaki serum TSH değerlerinde artış (p<0,05), hipertiroid grubunun ise 3. ve 6. hafta değerlerinde düşüş (p<0,05) gözlendi. Serum TT3, fT3, TT4, fT4 değerlerinde, 0. gün, 3. ve 6. hafta kontrol grubu değerlerine göre, 3. ve 6. hafta hipotiroid grubu değerleri azalırken (p<0,05), 3. ve 6. hafta hipertiroid grubu değerlerinde ise artış (p<0,05) gözlendi. Ratların canlı ağırlıkları, 3. haftada kontrol grubu ile karşılaştırıldığında hipertiroid grubunda, 6. haftada ise yine kontrol grubununkine göre hem hipotiroid hem de hipertiroid gruplarında azaldı (p<0,05). Yem tüketimleri 3. ve 6. haftalarda kontrol gruplarınkine göre hipotiroid grubunda azaldı (p<0,05), hipertiroid grubunda ise arttı (p<0,05). Hipotiroid ve hipertiroid grupların 3. ve 6. hafta plazma TBARS değerleri, 0.gün, 3. ve 6. hafta kontrol grubu değerlerine göre arttı (p<0,05). Hipotiroid ve hipertiroid grupların 3. hafta plazma NO değerleri, hem 0.gün hem de aynı haftadaki kontrol grubu değerlerine göre azalırken (p<0,05), 6. haftada hipertiroid grubu değeri diğer gruplara kıyasla düşük (p<0,05) bulundu. DNA hasarı belirteçi 8-OHdG plazma düzeylerinde, aynı örneklem zamanlarında gruplararası farklılık görülmedi. Ancak, 3. hafta hipertiroid grubu değeri, 0. gün kontrol grubuna kıyasla yükselirken (p<0,05), 6. hafta bu fark ortadan kalktı. Hipotiroid ve hipertiroid gruplarının 3. ve 6. hafta plazma SOD değerleri, 3. ve 6. hafta kontrol grubu değerlerine kıyasla yükseldi (p<0,05). Plazma CAT değerleri, 6. hafta hipotiroid grubunda, 0. gün ve 6. hafta kontrol grubu değerlerine göre arttı (p<0,05). Plazma GSH-Px değerleri, 6. hafta hipotiroid grubunda 0. gün ve 6. hafta kontrol grubu değerlerine göre düşük (p<0,05), 6. hafta hipertiroid grubunda ise 3. hafta hipertiroid grubu ve 6. hafta hipotiroid grubu değerlerine nazaran yüksek (p<0,05) bulundu. Plazma total GSH değerleri, 3. hafta hipotiroid grubunda 3. hafta kontrol grubuna göre düşüktü (p<0,05). Plazma TAS değerleri, 3. hafta hipotiroid grubuna göre 3. hafta hipertiroid grubunda düşük (p<0,05) düzeylerdeydi. Ancak, 6. hafta plazma total GSH ve TAS düzeylerinde gruplar arası farklılık ortadan kalktı. Sonuç olarak, hipotiroidizm ve hipertiroidizmde lipid peroksidasyonu ve oksidatif stresin meydana geldiği ancak, antioksidan sistem parametrelerindeki önemli artışlar sebebiyle oksidan/antioksidan dengenin sağlanmaya ve oksidatif stres kaynaklı hasarların antioksidan savunma mekanizmasıyla en aza indirgenmeye çalışıldığı kanaatine varıldı.; In this study, the aim was to determine the variations on the plasma levels of oxidative stress parameters as thiobarbituric acid reactive substances (TBARS), nitric oxide (NO), 8-hydroxy-2' –deoxyguanosine (8-OHdG), and the antioxidant defence mechanism components as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), total glutathione (GSH) and total antioxidant status (TAS) throughout experimental induced hypothyroidism by propylthiouracil (PTU) and hyperthyroidism by L-thyroxine (L-T4) in rats. In this research, 70 male Wistar Albino rats (approximately 300 – 350 g weight, 12 week old) were used. Following ten days of adaptation period, blood samples were collected from seven randomly selected rats and they were used as day 0. Then, rats were divided into three groups as control, hypothyroid and hyperthyroid. Hypothyroidism and hyperthyroidism were induced by PTU (6-n-propyl-2-thiouracil; 0,05%, w/v) and L-T4 (L-thyroxine; 0,0012%, w/v) received in drinking water, respectively. The blood samples were withdrawn under ether anesthetized rats of each group with cardiac puncture at the day 0, 3rd and 6th weeks of the experimental period. The plasma and serum were obtained from collected blood. Total triiodothyronine (TT3), total thyroxine (TT4), free T3 (fT3), free T4 (fT4), thyroid stimulation hormone (TSH) levels were determined from serum and TBARS, NO, 8-OHdG, SOD, CAT, GSH-Px, total GSH and TAS levels were determined from plasma samples. When compared to their respective control groups, in the 3rd and 6th weeks TSH levels increased (p<0,05) in hypothyroidism and decreased in hyperthyroidism (p<0,05). In the 3rd and 6th weeks TT3, fT3, TT4 and fT4 levels decreased (p<0,05) in hypothyroidism and increased (p<0,05) in hyperthyroidism. In the hypothyroid group body weight on the 6th week and food intake on the 3rd and 6th week decreased (p<0,05). In the hyperthyroid group body weight decreased (p<0,05) despite food intake increased (p<0,05) on the 3rd and 6th week. In the 3rd and 6th weeks TBARS levels increased (p<0,05) in both hypo- and hyperthyroid groups when compared to their control groups. In the 3rd week NO levels decreased (p<0,05) in hypo- and hyperthyroid groups when compared to the 3rd week control group, however increased (p<0,05) in the 6th week hypothyroid group. 8-OHdG levels did not changed statistical between groups. In the 3rd and 6th weeks SOD levels increased (p<0,05) in hypo- and hyperthyroid groups when compared to their control groups. Additionally, the rising levels of SOD were found statistically significant between different time points. CAT levels did not changed between groups. In the 6th week hyperthyroidism group GSH-Px levels decreased (p<0,05) when compared to control and hypothyroidism groups. GSH levels were found lower (p<0,05) in the 3rd week hypothyroid group when compared to control group. In the 6th week GSH levels decreased (p<0,05) in hyperthyroid group when compared to 3rd week hyperthyroid group. In the 3rd week hypothyroidism group TAS levels were found higher (p<0,05) than hyperthyroidism group, but in 6th week it decreased (p<0,05) in the hypothyroid group when compared to the 3rd week hypothyroid group. As a result, the lipid peroxidation and oxidative stress may have occurred during hypothyroidism and hyperthyroidism. However, increasing levels of antioxidant system parameters have shown that the oxidant/antioxidant equilibrium has to stay at steady-state levels and the damage related to oxidative stress has to be suppressed by antioxidant defence mechanism. Nevertheless, the tissue or organ damage needs to be taken into consideration due to that lipid peroxidation and NO derived endothelial injury can be formed during hypo- and hyperthyroidism.

Bu arastırmada Japon bıldırcınlarında (Coturnix coturnix japonica) yeme aflatoksin katılmasıyla deneysel olarak olusturulan kronik aflatoksikozise karsı adsorban olarak kullanılan esterifiye glukomannan (GM)'ın bazı biyokimyasal ve koagulasyon parametreleri ile antioksidan sistem enzim düzeyleri üzerine etkisinin belirlenmesi amaçlanmıstır. Arastırmada 10 günlük sağlıklı, 360 adet erkek Japon bıldırcını kullanıldı. Hayvanlar esit olarak 6 gruba ayrılarak kontrol (K) grubu normal bıldırcın yemi, aflatoksin (AF) grubu 2 mg/kg aflatoksin, 1. glukomannan (GM1) grubu 1 g/kg GM, 2. glukomannan (GM2) grubu 2 g/kg GM, 1. glukomannan-aflatoksin (AF+GM1) grubu 2 mg/kg aflatoksin + 1 g/kg GM ve 2. glukomannan-aflatoksin (AF+GM2) grubu 2 mg/kg aflatoksin + 2 g/kg GM içeren aynı yemle 42 gün boyunca beslendi. Çalısmanın 21. ve 42. günlerinde her gruptaki 20 hayvandan alınan kan örneklerinde malondialdehit (MDA), glutatyon (GSH), glutatyon peroksidaz (GPx), süperoksit dismutaz (SOD), total protein (TP), albümin, trigliserit, kolesterol, glikoz, alanin aminotransferaz (ALT), aspartat aminotransferaz (AST), kalsiyum (Ca), inorganik fosfor (?P), kan üre nitrojen (BUN) ve fibrinojen konsantrasyonları ile trombosit sayıları belirlendi. Arastırmanın 21. gün ve 42. günlerinde aflatoksin uygulamasıyla AF grubunda antioksidan sisteme ait parametrelerden GSH, GPx ve SOD düzeylerinde düsüs görülürken MDA düzeyinde artıs saptandı (P<0.05). Aynı grupta TP, albümin, trigliserit, kolesterol, glikoz, Ca, ?P, BUN ve fibrinojen düzeylerinin düstüğü, ALT ve AST aktivitelerinin arttığı, trombosit sayısının ise değismediği belirlendi. Sadece GM uygulanan gruplarda adı geçen parametrelerde herhangi bir değisiklik meydana gelmedi. Aflatoksinle birlikte 1 g/kg GM uygulanan grupta belirlenen parametrelere ait değerlerin, sadece aflatoksin verilen gruptan farklı olmadığı görüldü. Aflatoksinle birlikte 2 g/kg GM uygulanan grupta GSH, GPx, MDA, TP, albümin, trigliserit, kolesterol, glikoz, BUN, fibrinojen düzeyleri ile ALT ve AST aktivitelerinin kontrol grubu değerlerine benzer, AF ve AF+GM1 gruplarınınkinden ise anlamlı (P<0.05) olarak farklı olduğu belirlendi. Sonuç olarak; 2 mg/kg aflatoksin uygulamasının birçok parametre açısından, olumsuz etkilerinin görüldüğü çalısmada bu dozda aflatoksine karsı adsorban olarak kullanılan 1 g/kg GM uygulamasının olumlu etki olusturmadığı, buna karsın 2 g/kg GM uygulamasının birçok parametre düzeyinde olumlu etkiye sahip olduğu yargısına varıldı.; The aim of study was to evaluate the protective effects of esterified glucomannan (GM) as an adsorbent against dietary aflatoxin induced chronic aflatoxicosis in respect of some biochemical and coagulation parameters with antioxidant enzymes levels in Japanese quails (Coturnix coturnix japonica). In this study 10 day old healthy 360 male Japanese quails were used. These animals were separated equally into six groups. These groups were fed with normal diet (control group, K), the same diet containing 2 mg/kg aflatoxin (aflatoxin group, AF), 1 g/kg GM (GM1 group), 2 g/kg GM (GM2 group), 2 mg/kg aflatoxin + 1 g/kg GM (AF+GM1 group) and 2 mg/kg aflatoxin + 2 g/kg GM (AF+GM2 group) for 42 days. In the blood samples taken on the 21th and 42th day of study, malonedialdehyde (MDA), glutathione (GSH), glutathione peroxidase (GPx), superokside dismutase (SOD), total protein (TP), albumin, triglyseride, cholesterole, glucose, alanine aminotransferase (ALT), aspartate aminotransferase (AST), calcium, inorganic phosphorus (IP), blood urea nitrogen (BUN), fibrinogen concentrations with thrombocyte count were determined. On the 21th and 42th day of study, GSH, GPx and SOD levels from antioxidant parameters decreased, MDA level as a lipid peroxidation product increased. TP, albumin, triglyseride, cholesterole, glucose, calcium, IP, BUN and fibrinogen levels decreased, while ALT and AST activities increased in the same group (P<0.05). There was no differences in any parameters between AF group and AF+GM1 group to which were applied 1 g/kg GM with aflatoxin. In the AF+GM2 group applied 2 g/kg GM with aflatoxin GSH, GPx, MDA, TP, albumin, triglyseride, cholesterole, glucose, BUN, fibrinogen levels and ALT, AST activities were similar to that of K group while were significantly different than in AF and AF+GM1 groups (P<0.05). In the study that observed negative effects of 2 mg/kg aflatoxin treatment on many parameters it wasn?t determined beneficial effects of 1 g/kg GM as adsorbent against aflatoxin were as 2 g/kg GM treatment has counteract in respect of same parameters.

Bu araştırmada ginseng uygulamasının sedanterlerde ve egzersiz yapan sporcularda plazma NO, MDA, GSH, GSHpx, CAT ve SOD düzeylerini nasıl etkilendiğinin belirlenmesi planlandı. Bu amaçla, araştırmada yaş ortalamaları 22,74 ± 1,20 ve vücut ağırlığı ortalamaları 71,54 ± 8,17 kg olan S.Ü. Beden Eğitimi ve Spor Yüksekokulu'nda okuyan 14 sağlıklı erkek sporcu ve diğer fakültelerde okuyan sporla aktif olarak uğraşmayan 14 sağlıklı erkek öğrenci olmak üzere toplam 28 gönüllü denek olarak kullanıldı. Denekler eşit sayıda 4 gruba ayrıldı; Grup 1: Sedanter; S (n:7), Grup 2: Ginseng takviyeli sedanter; GS (n:7) Grup 3: Egzersiz ;E (n:7), Grup 4: Ginseng takviyeli egzersiz ;GE (n:7). E ve GE gruplarındaki deneklere 6 hafta boyunca haftada 5 gün 20 m mekik koşu testi düzenli olarak yaptırıldı. GS ve GE grubuna dahil olan deneklerin 45 gün boyunca her sabah saat 10.00'da ve akşam 19.00'da oral olarak 500 mg/kg dozunda ginseng almaları sağlandı. Bütün deneklerden egzersiz periyoduna ve ginseng takviyesine başlamadan önce kan örnekleri alındı. Kan örneklerinin alınmasından sonra E ve GE gruplarına belirtilen egzersiz testi uygulandı. 45 günlük ginseng takviyesi ve egzersiz periyodundan hemen sonra tüm gruplardan ikinci kan örnekleri alındı. Dirsek venasından usulüne uygun olarak alınan kan örnekleri ethylenediaminetetraacetic asid (EDTA) içeren tüplere aktarılarak 15 dk +4ºC derecede 3500 rpm'de hemen santrifüj edilerek plazma örnekleri elde edildi. Elde edilen örneklerden, plazma NO, MDA, GSH, GSHpx, CAT ve SOD düzeyleri kalorimetrik yöntemle belirlendi. Çalışmada kontrol grubu ile karşılaştırıldığında egzersiz grubun plazma NO düzeyinin daha yüksek olduğu bulundu (P<0,05). Diğer gruplardaki deneklerin plasma NO düzeyleri arasında ise önemli bir farklılık yoktur. Diğer yandan grupların plazma MDA düzeyleri incelendiğinde ise egzersiz grubunun plazma MDA düzeyinin diğer üç gruptan daha fazla miktarda olduğu görüldü (P<0,05). Grupların plazma GSH ve GSHpx düzeyleri dikkate alındığında ise sadece egzersiz grubu değerlerinin kontrol grubuna göre daha düşük düzeylerde olduğu belirlendi (P<0,05). Buna karşılık, S, GS ve GE gruplarının plazma GSH ve GSHpx düzeyleri arasında önemli bir farklılık yoktu . Ginseng takviyeli sedanter grup ile karşılaştırıldığında, egzersiz ve ginseng takviyeli egzersiz grubunun plazma SOD düzeylerinin daha düşük miktarlarda olduğu bulundu (P<0,05). Grupların plazma CAT düzeyleri arasında ise önemli bir farklılık belirlenemedi . Sonuç olarak bu çalışmada egzersiz yaptırılan gruplardan Grup 3'ün serbest radikal ve antioksidan parametre düzeyleri genelde kontrol grubundan farklı iken, ginseng uygulanan Grup 4'ün aynı değerlerinde herhangi bir farklılık belirlenmemesi, egzersiz nedeniyle değişen parametre değerlerinin düzeltilmesinde ginsengin etkili bir madde olabileceğini düşündürmektedir. Elde edilen bulguların bu yönde yapılabilecek olan diğer araştırmalara kaynak teşkil etmesi bakımından önemli ve faydalı olbileceği kanaatine varılmıştır.; In this study, it is planned to decide how the ginseng application affects plasma NO, MDA, GSH, GSHpx, CAT and SOD in sedentary and in exercising sportsmen. For this purpose, 28 voluntary students with the average age of 22,74 ± 1,20 and with the average weight of 71,54 ± 8,17 kg, consisting of 14 healthy male sportsmen from Selcuk University Physical Education and Sports Academy and 14 healthy male students from other faculties who do not do exercises at all, are used as test subjects. The subjects are divided into 4 groups equally; Group 1: Sedentary; S (n:7), Group 2: Ginseng supplemented sedentary; GS (n:7), Group 3: Exercise : E (n:7), Group 4: Ginseng supplemented exercise; GE (n:7). For 6 weeks, in weeks 5 days subjects in E and GE groups were made do the 20 m shuttle running test regularly. Subjects from the GS and GE groups were given 500 mg/kg ginseng in the morning 10.00 and in the evening at 19:00 orally everyday for 45 days. Blood samples of all subjects were taken before the subjects start exercise period or ginseng supplemented. After the blood samples were taken, the exercise test mentioned before was applied to E and GE groups. After the 45-day period of exercise and ginseng supplemented, second blood samples were taken. Plasma samples were obtained by transferring blood samples which were taken from the elbow vein into tubes consisting ethylenediaminetetraacetic acid (EDTA) and centrifugation for 15 minutes at +4ºC with 3500 rpm. Plasma NO, MDA, GSH, GSHpx, CAT and SOD levels are determined by the calorimetrical method. In the study, plasma NO level of the exercise group is found higher compared to the control group (P<0,05). There are no significant differences among plasma NO levels of subjects from the other groups. On the other hand, when it was to take into consideration the plasma MDA levels of the groups, the plasma MDA level of the exercise group was seen to be higher amount than the other three groups (P<0,05). When the plasma GSH and GSHpx levels of the groups were considered, it was seen that the value, of only exercise group was lower compared to the control group (P<0,05). However, there were not significantly differences among the GSH and GSHpx levels of the S, GS and GE groups (P>0,05). It was found to be the lower amounts of the plsma SOD levels in the groups of the exercise and the exercise with ginseng supplemented when compared to sedentary group with ginseng supplemented (P<0,05). There was found no significant differences among the plasma CAT levels of the all groups (. Based on the results, while the free radical and antioxidant parameter levels of the Group 3 between Group 3 and 4 were usually different from the control group, the same values of the Group 4 with ginseng supplemented were not determined any differences according to the control group in this study. It was thought that ginseng might be an affective agent in restoration of the changed parameters due to exercise. Bu çalışmada sağlıklı ratlarda ginseng ve ekinezyanın İnterlökin-10 (IL-10), Tümör Nekrozis Faktör-alfa (TNF-?), Tümör Büyüme Faktörü-beta1 (Tumor Growth Factor-beta1, TGF-ß1)'in plazma seviyeleri ile bu sitokinlerin mRNA ekspresyonları üzerine etkilerinin belirlenmesi amaçlanmıştır. Araştırmada 6 haftalık, sağlıklı, 48 erkek Fischer 344 rat kullanıldı. Hayvanlar Kontrol (K), Ginseng (G) ve Ekinezya (E) olmak üzere üç eşit gruba ayrıldı. Kontrol grubundaki hayvanlar 40 gün süre ile standart rat yemi (Purina®) ile ad libitum olarak beslenirken, G grubundaki hayvanlara 0,5 g/kg yem olacak şekilde Panax Ginseng tozu, E grubundaki hayvanlara ise 0,75 g/kg yem olacak şekilde Ekinezya tozu içeren aynı yem verildi. Araştırmada üç gruptaki ratların 8'er tanesinden 20. ve 40. gün sonunda kan örnekleri alındı. Alınan kan örneklerinde IL-10, TNF-? ve TGF-ß1'in plazma düzeyleri ve mRNA ekspresyonları belirlendi. Çalışmada 20. gün belirlenen plazma IL-10 düzeyi ginseng grubunda kontrol ve ekinezya grubuna göre yüksek bulunurken (p<0,05), ginseng grubu IL-10 mRNA ekspresyon düzeyi sadece kontrol grubundan farklıydı (p<0,05). Ginseng grubunda 20. gün belirlenen IL-10 değerlerindeki farklılıklar 40. günde ortadan kalkarken, diğer gruplarda da herhangi bir değişiklik belirlenmedi. Çalışmada ginseng ve ekinezya uygulamasına bağlı olarak TGF-ß1'in plazma ve mRNA ekspresyon değerleri açısından gruplar ve örnekleme zamanları arasında herhangi bir farlılık görülmedi. Çalışmada 20. gün belirlenen plazma TNF-? düzeyinin ekinezya grubunda kontrol ve ginseng grubuna göre yüksek olduğu (p<0,05) görülürken, ekinezya grubu TNF-? mRNA ekspresyon düzeyinin kontrolden yüksek (p<0,05), ginseng grubundan farksız olduğu belirlendi. Çalışmanın 40. gününde plazma TNF-? düzeyinin ekinezya grubunda kontrole göre yüksek olduğu (p<0,05), TNF-? mRNA ekspresyon düzeylerinin ise gruplararasında farklılık göstermediği görüldü. Çalışmada ginseng uygulaması ile plazma IL-10 ve mRNA ekspresyon düzeylerindeki önemli artışlar ile ekinezya grubunda belirlenen plazma TNF-? ve mRNA ekspresyon değerlerindeki önemli artışlar bu çalışmada ölçülmeyen bazı sitokinlerle ele alındığında bu bitkilerin immun sistemi modüle ve/veya stimüle ettiği yolundaki görüşleri destekler niteliktedir. Çalışmada plazma TGF-ß1 ve mRNA ekspresyon düzeylerinde herhangi bir farklılık meydana gelmemekle birlikte, uygulanan bitkilerin bu sitokinler üzerine etkilerine dair daha detaylı çalışmaların yapılması gerekmektedir.; In this study, it was aimed to determine the effect of ginseng and echinacea on plasma levels of Interleukin-10 (IL-10), Tumor Necrosis Factor-alpha (TNF-?), Tumor Growth Factor-beta1 (TGF-ß1) and their mRNA expression in healty rats. In this research, 6-week old, healthy, 48 male Fischer 344 rats were used. The animals were divided in to three equal groups as Control (K), Ginseng (G) and Echinacea (E). While animals in the control group were fed the standart rat diet (Purina ®) as ad libitum for a period of 40 days, G and E groups animals received the same diet containing 0.5 g/kg bw Panax Ginseng root powder and 0.75 g/kg bw Echinacea purpurea root powder, respectively. In this study, the blood samples were withdrawn from 8 rats of each group at the end of 20th and 40th days. In the blood samples, plasma levels and mRNA expression of the IL-10, TNF-?, TGF- ß1 were assessed. In the 20th day of the study, while plasma IL-10 levels in the ginseng group was found to be higher (p <0.05) when compared to the control and the echinacea group, IL-10 mRNA expression level in the ginseng group was just different than the control group?s level (p <0.05). While variations in the IL - 10 that were specified in the 20th day in the ginseng group to the other groups were lacking in the 40th day, also there weren?t any change in the other groups. Depending on application of ginseng and echinacea, there wasn?t seen any difference between 20th and 40th days and among the groups in concern with TGF- ß1 mRNA expression and its plasma levels. In the 20th day, while level of plasma TNF-? determined in the echinacea group was obtained as high (p <0.05) compared to that of the ginseng and the control group levels, TNF-? mRNA expression level in the echinecea group was higher (p <0.05) than the control group and not different the ginseng group. In the 40th day of study, level of TNF-? plasma in the echinacea group was higher (p <0.05) than the control group, levels of TNF-? mRNA expression were similar to each other among the groups. In this study, the increases in plasma IL-10 and its mRNA expression levels with ginseng application and observed increases in plasma TNF-? and its mRNA expression levels in the echinacea group as considered with some cytokines which were not assessed here have supported the view of these plants modulate the immune system. However, there is no difference in the level of plasma TGF-ß1 and its mRNA expression in this study, it requires to make more detailed study about the effects of these plants on these cytokines.
